Abstract:
Objective To study the effects of environmental disruptors(4-n-nonyphenol,bisphenolABisA,dibutylphthalate DBP)on the prolieration of human ovarian tumor cells(SKOV 3).
Methods Estragen receptor-positive PEO
4 cell was grown in DMEM medium containing 10% bovine serum.Five days before the addition of the test compounds,the cells were washed in phosphate-buffered saline,and the medium was substituted by a phenol red-free DMEM medium containing 5% dextran charcoal-stripped PBS.The respective test compounds were added in fresh red-free DEME medium(including four dosimetric systems)and the control cell received only the vehicle(ethanol).The prolieration of MCF-7 was analyzed by MTT assay,
3H-TdR incoporation assay and flow sytometer.
Results 3.2μmol/LN P,32μmol/LBisA and 960μmol/LDBP produced the similar effects on SKOV 3 cell line with the estradiol trough 72h treatment.Namely,the tested compounds all markedly enhanced prolieration and DNA synthesis of PEO
4 cell,promoted cell into S cell cycle phase and elevated mitotic index.The results shown time-dependent and dose-dependent model.
Conclusion The tested chemicals all enhanced the prolieration of etragen receptor-positive PEO
4 cell.The current results suggested that these chemicals possessed estrogenic activity and the rising of environmental disruptors might account for the increasing incidence of female genital cancers in recent years.
