氧化苦参碱防治肝纤维化作用

Effect of oxymatrine on hepatic fibrosis

      目的   观察氧化苦参碱的抗肝纤维化作用并探讨其作用机制。
      方法   用CCl4诱导昆明小鼠建立肝纤维化模型, 设正常对照组、模型组、氧化苦参碱预防组和治疗组。实验结束后分别测定丙氨酸氨基转移酶、白蛋白、总胆红素、透明质酸、层粘连蛋白、Ⅲ型前胶原、Ⅳ型胶原, 并作肝组织病理学形态分析及免疫组化染色。
      结果   氧化苦参碱预防组和治疗组与模型组比较, 肝功能、肝纤维化指标、病理学形态分析及免疫组化染色结果差异均有统计学意义(P < 0.05), 且预防组与治疗组比较, 差异有统计学意义(P < 0.05)。
      结论   氧化苦参碱通过抑制肝内胶原合成、减少结缔组织生长因子的表达, 抑制肝脏细胞外基质异常增生发挥抗肝纤维化作用。

     

    Abstract:
      Objective   To observe the antihepatofibrosis function of oxymatrine and to discuss the mechanism.
      Methods   this experiment is based on the model of kunming mouse hepatic fibrosis(CCl4), and normal group, model group, oxymatrine prevent group and therapy group were set up.After that, ALT, ALB, Tbil, HA, LN, PC-Ⅲ, Ⅳ-CL were measured separately, simultaneously hepatic tissue pathology and immunohistochemistry dyeing were used.
      Results   Oxymatrine prevention group and therapy group were more distinct than those of model group on hepatic function, the indexs of hepatic fibrosis, pathology changes and the result of immunohistochemistry dyeing(P < 0.05).And compared with therapy group, prevention group had markedly difference(P < 0.05)in those indexes.
      Conclusion   Oxymatrine can restrain the production of collagen, decrease the expression of CTGF and inhibit the abnormal hyperplasia of hepatic extracellular matrix, so ithas the function of antihepaticfibrosis.
