白术提取液安全性评价

Safety assessment of Baizhu

      目的   评价白术提取液的安全性.
      方法   急性经口毒性评价采用最大耐受量法, 细胞毒性评价采用噻唑蓝(MTT法)、中性红吸收法(NPI法)和集落形成法, 遗传毒性评价采用小鼠骨髓嗜多染红细胞微核试验、Ames试验、小鼠淋巴细胞杀伤细胞(TK)基因突变试验和彗星试验.
      结果   白术提取液急性经口毒性评价属实际无毒级物质; MTT法和中性红吸收法在受试剂量范围内未观察到对细胞的损伤作用, 集落形成法在2.5~10.0 g/L剂量范围内表现出剂量依赖性的细胞毒性; 4种遗传毒性试验均为阴性结果, 即无遗传毒性.
      结论   白术水提物属于一种低毒性物质.

     

    Abstract:
      Objective   To assess the safety of Baizhu(Atractylodes macrocephala Koidz).
      Methods   Maximal tolerance test was used to assess acute oral toxicity; methyl thiazolyl tetrazolium(MTT)test, neutral red intake test and clone-forming test were used to assess cytotoxicity; micronucleus test of bone marrow cell, Ames test, mouse lymphoma TK gene mutation assay and comet assay were used to assess genotoxicity.
      Results   The extract of Baizhu was nontoxic according to the acute toxicity test, In MTT test and neutral red intake test, cytotoxicity was not observed at the doses tested, in clone-forming test the dose-dependent cytotoxicity was obser ved at the dose range of 2.5~10.0 g/L.The results of four genotoxicity tests were all negative, which showed there was no genotoxicity.
      Conclusion   The water-soluble extract of Baizhu is of low toxicity.
