Effect of SDS on resonance light-scattering spectrum of histone and DNA complex

  • 摘要:
      目的   研究十二烷基磺酸钠(SDS)对组蛋白与DNA共振光散射强度的影响, 探讨三者相互作用规律.
      方法   pH 7.5的Tris-HCl缓冲液体系中, 将组蛋白与DNA及SDS混合.在入射光波长和荧光波长相同条件下, 用荧光分光光度计测定混合液的共振光散射强度.
      结果   扫描光谱显示最大光散射波长为302nm, 最佳pH值为7.5, SDS浓度在0.02~0.15mmol/L范围内, 体系共振光散射强度达到最大值且趋于稳定.
      结论   小剂量SDS能增强组蛋白-DNA共振光散射体系的光散射强度, 反之, 大剂量SDS促使组蛋白与DNA分离, 使光散射强度大大减弱.

     

    Abstract:
      Objective   To study the effect exerted by sodium dodecyl sulfate(SDS)on the resonance lig h-tscatter ing based on histone and DNA complex and to explore the orderliness of inter action in the three compounds.
      Methods   Histone, SDS and DNA were mix ed into the Tris-HCl buffer solution(pH 7.5).Common spectrofluoromenter was used to measure the resonance light-scattering intensity of the mix ture with same excitation and emission waveleng th.
      Results   Scan spectrum show ed that the max imal light-scattering wavelength was 302nm, and the optimum measurement for ligh-tscattering intensity was at pH 71 5.The ligh-tscattering sig nal was strong in the range of 0.02~0.15 mmol/LSDS.
      Conclusion   Lo w dose of SDS improv ed the resonance ligh-tscattering intensity of histone and DNA.Otherwise, high dose of SDS urged the segregation of histone and DNA, socut down thereso nance light-scatter ing intensity greatly.
