Effect of zinc on apoptosis and P53 et al gene expression in HL-60 cells

      目的   研究微量元素锌在体外对HL-60细胞凋亡的作用机制.
      方法   HL-60细胞常规培养24 h, 分为对照组、四吡啶甲基乙二胺N, N, N', N'-tetrakis(2-pyridylmethyl)ethylenediamine(TPEN)处理组、TPEN+ZnSO 4组.以细胞形态学改变、流式细胞仪和DNA凝胶电泳分析为凋亡观察指标, 并用mRNA狭缝杂交和Western-blot测定细胞凋亡相关基因的改变.
      结果   细胞内Zn 2+减少可诱导人髓性细胞白血病细胞系HL-60凋亡, 引起bcl-2、c-mycmRNA及蛋白的表达降低, 补充ZnSO 4(终浓度为20μmol/L)可改善上述现象.
      结论   细胞内Zn 2+减少诱导HL-60细胞凋亡过程与细胞内下调bcl-2、c-mvc基因的表达有关.

     

    Abstract:
      Objective   To study the effect of tr ace element zinc on apoptosis in HL-60 cells and its mechanism.
      Methods   The HL-60 cells were cultured by usual rules for 24 h and then divided into group of control, N, N, N., N.-tetrakis(2-pyridylmethyl)ethylenediamine(TPEN).TPEN+ZnSO 4.The apoptosis of cells w as observed by means of morphological changes, flow cytometry analysis and agarose gel electrophoresis of DNA.The genes expression asso ciated with apoptosis were detected in both mRNA and protein levels by mRNA slo-tblot and western-blot.
      Results   Intacellular Zn 2+ decrease could induce apoptosis in HL-60 cells and dow n-regulate expression of genes bc-l 2, c-myc in both mRNA and protein levels, but additiion of ZnSO 4(final concentration of 20 Lmol/L)improved the conditions.
      Conclusion   Down-regulated expression of genes bc-l 2, -cmyc are associated with the process of apoptosis induced by intracellular Zn 2+ decrease.
