茶多酚对人肝癌细胞激活蛋白1及细胞周期影响

Effect of tea polyphenols on cell cycle of human liver cancer cell

  • 摘要:
      目的   观察茶多酚对人肝癌细胞激活蛋白1(AP-1)及细胞周期的影响。
      方法   构建AP-1报告质粒测量AP-1活性, 免疫组化测量细胞周期素D1和细胞周期素依赖激酶4表达变化, 流式细胞仪分析细胞周期。
      结果   茶多酚抑制AP-1活性, 细胞周期素D1和细胞周期素依赖激酶4表达降低, 细胞阻滞在G1期。
      结论   茶多酚可能通过下调AP-1活性使细胞周期素D1表达降低; 而茶多酚使细胞周期素依赖激酶4表达降低, 可能与AP-1的活性无关。

     

    Abstract:
      Objective   To study the effects of tea polyphenols on activator protein 1 and cell cycle of human liver cancer cell.
      Methods   An activator protein 1 luciferase reporter gene was constructed and the activities of activator protein 1 were measured.The expression changes of cyclin D1 and cyclin-denpendent kinase 4 were studied by the immunity organization chemistry method.cell cycle was analyzed by flow cytometry.
      Results   The activities of activator protein 1 were repressed, the experession of cyclin D1 and cyclin-denpendent kinase 4 was reduced, G1 arrest was induced by tea polyphenols in cell line.
      Conclusion   Tea polyphenols possibly repressed the expression of cyclin D1 through down-regulating the activities of activator protein 1;It had nothing to activities of activator protein 1 that tea polyphenols repressed the expression of cyclin-denpendent kinase 4.
