Prediction and production of epitope of SARS-CoV

  • 摘要:
      目的   获得严重急性呼吸道综合征(SARS)冠状病毒(CoV)中和性抗原蛋白.
      方法   使用Blast工具将SARSS蛋白的氨基酸序列与其他蛋白进行比对分析.冠状病毒鼠肝炎病毒的刺突蛋白在S2段存在中和性抗体表位.通过对鼠肝炎病毒和SARSCoVS2的氨基酸序列分析, SARSCoV的S2段类似于鼠肝炎病毒的S2段.使用DNASTAR软件对2段的相似性进行分析.选取SARSCoVS2蛋白基因在大肠埃希菌中进行表达.表达产物进行免疫印迹分析.
      结果   在SARS刺突蛋白C端发现了与其他冠状病毒S2蛋白具有较高的同源性的片段.在SARS病毒刺突蛋白的S2段可能存在中和性表位.将该肽段在大肠埃希菌中获得表达.表达的蛋白片段可以和来自SARS恢复期的人抗SARS-CoV免疫球蛋白特异性结合.
      结论   表达的蛋白片段有可能成为预防SARS的候选疫苗.

     

    Abstract:
      Objective   To obtain the peptide of severe acute respiratory syndro me(SARS)-CoVcontained the neutralizing Bcell epitope.
      Methods   The amino acid sequence of majorst ructure protein on the outermembrane of SARS-CoVwas analyzed by blast with known pro tein.The coronvirus mouse hepatitis virus(MHV)contained a neutralizingepitope in the S2 region.The SARS-CoVcontained the same region with MHVin the S2 segment.The amino acid sequence of the neutralizingepitope region of SARS-CoVand MHVwas analy zed with the DNASTARsoftware.The gene of the peptide contain the selected region was expressed in E.coli.The expressed peptide was identified by Western-blotting.
      Results   Aconserved seg ment which named S2 seg ment of coronvirus was digoutin the C-terminal of the spike protein of SARS-CoV.The SARS-Co Vmay contained a neutralizingepitope in the S2 region.The gene contained the selected region was expressed in E.coli.The expressed peptide could be reco gnized by the human anti-SARS-CoVantibody collected from the convalescence patients infected SARS.
      Conclusion   The expressed peptide may be a selected vaccine forprevention of SARS.
