硒对糖尿病大鼠肝脏蛋白磷酸酶基因表达影响

Effects of selenium supplementation on protein phosphatase 2A expression of liver in diabetic rats

      目的   研究补硒对糖尿病大鼠肝脏代谢相关基因表达的影响。
      方法   通过对前期研究分离到的与补硒有关的糖尿病大鼠差异显示的基因片段进行克隆、测序, 并进行同源性分析。根据待检测的基因序列设计引物, 进行RT-PCR检测, 以观察各组大鼠之间肝脏中基因表达的变化。
      结果   经过克隆、测序、同源性比较, 差异显示片段Se-8的碱基序列与蛋白磷酸酶2A(protein phosphatase 2A, PP2A)的同源性为99%。糖尿病对照组(0.507 2±0.057 4)、糖尿病补硒组(0.698 2±0.039 6)PP2A表达量低于正常对照组(1.330 3±0.185 5)(P < 0.05);糖尿病补硒组PP2A的表达量(0.698 2±0.039 6)高于糖尿病对照组(0.507 2±0.057 4)(P < 0.05)。
      结论   补硒对糖尿病大鼠肝脏PP2AmRNA表达有上调作用, 这可能是硒改善糖尿病糖、脂代谢紊乱的分子机制之一。

     

    Abstract:
      Objective   To study the effects of selenium supplementation on related gene expression of liver in diabetic rats.
      Methods   cDNA fragments from former research project were cloned, sequenced and BLASTn analysed.The RT-PCR were made using the primers designed acco rding to the sequence of cDNA.
      Results   The identities between the sequence of Se-8 and protein phosphatase 2A(PP2A)was 99%.The expression level of DM group(0.507 2±0.057 4)and DM+Se group(0.698 2±0.039 6)was obviously lower than those of NC group(1.330 3±0.185 5)(P < 0.05).The ex pression level of DM+Se group(0.698 2±0.039 6)had some improvement compared with those of DM group(0.507 2±0.057 4)(P < 0.05).
      Conclusion   Selenium supplementation could improve the expression of PP2A mRNA on liver in diabetic rats.It may be one of the molecular mechanisms that Selenium could improve the disorders of glucose and lipids metabolism in diabetic rats.
