铅对小鼠睾丸TGFβ1及Caspase-3表达的影响

Effects of lead on expressions of TGFβ1 and Caspase-3 in mice testes

      目的   探讨不同浓度和时间醋酸铅染毒对小鼠睾丸转化生长因子β1(Transforming growth factorβ1, TGFβ1)、半胱天冬酶3(Caspase-3)表达的影响, 进一步阐明醋酸铅所致睾丸毒性的作用机制。
      方法   45只雄性小鼠分为空白对照组和0.2%, 0.4%醋酸铅染毒组, 每组15只。并在第2, 4, 6周分别处死每组小鼠各5只, 取其睾丸组织, 免疫组化检测TGFβ1、Caspase-3表达的差异。
      结果   染铅后不同周TGFβ1、Caspase-3的表达较空白对照组增强, 差异有统计学意义(P < 0.05)。且随着染毒时间的延长, 表达越强(P < 0.05)。但在0.2%与0.4%不同周染铅组之间TGFβ1、Caspase-3的表达差异并无统计学意义(P > 0.05)。
      结论   醋酸铅使睾丸TGFβ1Caspase-3的表达增强, 从而促使生殖细胞发生凋亡。

     

    Abstract:
      Objective   To ex plore the effects of ex pressions of TGFβ1, Caspase-3 in mice testes at different concentrations and time of lead acetate, and further to elucidate the toxic mechanism.
      Methods   Forty five mice were randomly divided into blank control group and 0.2%, 0.4% lead acetate exposed group(Fifteen mice in each group).Five mice in each group were put to death at two, four and six weeks respectively and then testes were separated from mice.Expressions of TGFβ1, Caspase3 were detected by immunohistochemistry.
      Results   The levels of TGFβ1 and Caspase-3 were significantly higher in the lead exposed groups than those in the control groups(P < 0.05).Furthermore, the levels increased along with the time of lead exposed increased(P < 0.05).But there were no significant difference between 0.2% and 0.4% lead exposed groups(P > 0.05).
      Conclusion   Lead acetate increases the expressions of TGFβ1 and Caspase-3, which induces apoptosis of germ cells.
