葡多酚对小鼠乳腺癌细胞增殖活性抑制作用

      目的   研究葡多酚(GPC)对小鼠乳腺癌细胞增殖活性的抑制作用。
      方法   将皮下接种乳腺癌(EMT 6)细胞株的小鼠每日经口灌胃给予不同剂量的GPC, 13 d后取乳腺癌组织, 用四甲基偶氮噻唑蓝(MTT)法测各组小鼠乳腺癌细胞的增殖活性, 免疫组化法检测各组增殖细胞核抗原(PCNA)和p 53表达。
      结果   肿瘤对照组PCNA和p53阳性表达率分别为72.78%和37.22%, 高剂量GPC组则分别为28.32%和7.46%, 差异有统计学意义(P < 0.05)。各组乳腺癌细胞的增殖活性随GPC剂量的增高而降低。
      结论   葡多酚可有效抑制小鼠乳腺癌细胞增殖活性。

     

    Abstract:
      Objective   To study the inhibition effect of grape procyanidin(GPC)on the proliferation of breast carcinoma cells.
      Methods   Different dose of GPC was given by mouth to the mice inoculated EMT 6 breast carcinoma cells.The proliferation of brease carcinoma cells was determined by methy 1 tetrazolium(MTT)assay, and the proliferation cell nuclear antigen (PCNA)and mutant p 53 protein expression of mice breast carcinoma cells was measured by immunohistochemistry method after 13 days.
      Results   The expression rates of RCNA and mutant p53 protein of the carcinoma control group were 721.28% and 371.22% respectively, and those of the high dose GPC group were 28.32%and 7.46%.The differences between the two groups were significant(P < 0.05).The proliferation of mice breast carcinoma cells offered reducing tendency with the increasig dose of GPC indifferent groups.
      Conclusion   GPC has an inhibition effect on the proliferation of the breast carcinoma cells.
