Effects of RNA interference on Bax gene on 293 cell apoptosis induced by cadmium

      目的   利用RNA干扰(RNAi)技术探讨Bax基因在镉致293细胞凋亡过程中的作用。
      方法   细胞转染siRNA-Bax后染镉, 用rt-PCR、Westernblotting法测Bax基因表达, 流式细胞术测细胞凋亡, 四甲基偶氮噻唑蓝(MTT)法测细胞活力。
      结果   30(mol/L氯化镉可使293细胞Bax基因mRNA和蛋白表达明显增高, 细胞凋亡增加。siRNA-Bax可抑制这些效应, 而错义siRNA-Bax和单纯转染试剂无效。
      结论   Bax基因在镉致293细胞凋亡过程中起到关键作用。

     

    Abstract:
      Objective   To study the effects of Bax in 293 cell apoptosis induced by cadmium chloride with RNAi.
      Methods   Cells were incubated with CdCl2 after siRNA transfection.The expressions of Bax were detected by RT-PCR and western blot, the rates of apoptosis were measured by flow cytometry(FCM)and the activities of cells were measured by MTT.
      Results   The apoptosis rates of 293 cells treated with 30LM CdCl2 were higher than those of the control group accompanied by increased expressions of Bax.siRNA-Bax could inhibit those changes but negative control and transfection agent couldn't.
      Conclusion   Bax is a key role during cadmium chloride induced 293 cells apoptosis.
