全氟辛烷磺酸对大鼠兴奋性氨基酸影响

Study on excitatory amino acids in PFOS-exposed rats by HPLC

  • 摘要: 目的探讨全氟辛烷磺酸(PFOS)与大鼠中枢神经系统兴奋性氨基酸的时间反应关系。方法将成年Wistar大鼠随机分为1个对照组和4个实验组25 mg/(kg·bw),染毒后2,4,8,24 h;用PFOS对成年Wistar大鼠经口1次染毒;对照组给予等体积的2%吐温-80溶液。高效液相色谱法(HPLC)测定脑组织中谷氨酸、天冬氨酸、γ-氨基丁酸及甘氨酸含量变化。结果谷氨酸2,4,8 h含量分别为(0.89±0.162),(0.75±0.276),(0.84±0.542)μmol/(g·protein),与对照组(1.30±0.063)μmol/(g·protein)比较,差异有统计学意义(P<0.05)。天冬氨酸无显著变化。甘氨酸8,24h含量分别为(1.24±0.352),(1.09±0.215)μmol/(g·protein),与对照组(1.54±0.120)μmol/(g·protein)比较,差异有统计学意义(P<0.05)。γ-氨基丁酸含量24 h为(1.68±0.188)μmol/(g·protein),与对照组(2.03±0.370)μmol/(g·protein)比较,差异有统计学意义(P<0.05)。结论低剂量PFOS染毒后中枢神经系统出现先抑制后兴奋的症状可能与兴奋性氨基酸的参与有关。

     

    Abstract: ObjectiveTo explore the time-response relationship between central neural excitatory amino acids(EAAs) and PFOS.MethodsWister rats were randomly divided into l control group and 4 duration2,4,8,24 h after 25mg/(kg·bw)exposuregroups.Wister rats were exposed once orally.The controls were treated with 2% tween-80.The contents of glutamate(Glu),aspartate(Asp),glycine(Gly)and gamma-aminobutyric acid(GABA)in brain tissues were determined by high performance liquid chromatography(HPLC).ResultsGlu content at 2,4,8 h were(0.89±0.162),(0.75±0.276), (0.84±0.542)μmol/(g·protein),and there was significant difference compared with corresponding control group(1.30±0.063)μmol/(g·protein)(P<0.05);Asp did not show evident changes;Gly content at 8,24h were(1.24±0.352), (1.09±0.215)μmol/(g·protein),and there was significant difference compared with corresponding control group(1.54±0.120)μmol/(g·protein)(P<0.05).GABA content at 24h were(1.68±0.188)μmol/(g·protein),and there was significant difference compared with corresponding control group(2.03±0.370)μmol/(g·protein)(P<0.05).ConclusionEAAs appears to be related with the development of inhibited activity and then excitatory symptom by the low dose of PFOS exposed.
