绿茶多酚对牛内皮细胞窖蛋白-1表达影响

Influence of green tea polyphenols on caveolin-1 expression in bovine endothelial cells

  • 摘要: 目的 探讨绿茶多酚(GTPs)对牛内皮细胞窖蛋白-1(Cav-1)表达的影响。方法 以体外原代培养的牛主动脉内皮细胞(BAECs)为研究对象;以4μg/ml的绿茶多酚分别作用于牛内皮细胞0,4,8,12,16和24 h,免疫印迹方法观察Cav-1的蛋白水平变化;RT-PCR检测窖蛋白-1的mRNA表达水平变化。结果 Cav-1蛋白和mRNA表达水平在4μg/ml GTPs作用4 h开始下降,可以维持到8 h,在12 h后Cav-1蛋白和mRNA恢复到正常水平。结论 绿茶多酚可降低内皮细胞窖蛋白-1的蛋白和mRNA表达,并且有时效性,可能是茶及其多酚改善内皮功能和预防心血管疾病的分子机制之一。

     

    Abstract: Objective To investigate the influence of green tea polyphenols(GTPs)on caveolin-1(Cav-1)expression in bovine endothelial cells in the level of cell and molecular initially.Methods In vitro,primary cultured bovine artery endothelial cells(BAECs)were treated with 4 μg/ml GTPs for 0,4,8,12,16 and 24 hours,respectively.After protein and RNA extracted from treated cells,the Cav-1 protein and mRNA levels were detected by Westem Blot and RT-PCR methods.Results After BAECs co-incubated with 4 μg/ml GTPs for 4 hours,the expression of Cav-1 protein and mRNA decreased.The down-regulation effect of GTPs can maintain 4 hours and protein and mRN A expression of Cav-1 regained to normal level after 8 hours.Conclusion GTPs can decrease the protein and mR NA expression of Cav-1 in endo thelial cells,and the influence of GTPs may be chronergy.It may be the molecular mechanism of tea amelioration endo thelial function and prevention CVD.
