漏芦对细胞色素P450酶活性及mRNA表达影响

Effects of rhaponticum uniflorum (L.) DC on activity and mRNA expression of cytochrome P450

  • 摘要: 目的 研究漏芦对CYP2E1酶活性及其mRNA表达水平的影响。方法 以大鼠原代肝细胞为研究对象,实验组分别加入不同浓度漏芦(15.6,31.2,62.4 mg/ml)处理48 h,用苯胺羟化酶(aniline hydroxylase,ANH)活性反映CYP2E1酶活性,采用逆转录聚合酶链反应(RT-PCR)检测CYP2E1mRNA表达水平。结果 漏芦作用后,CYP2E1酶活性及相应mRNA的表达呈浓度依赖性抑制。漏芦在15.6,31.2,62.4 mg/ml浓度下对CYP2E1酶活性的抑制率分别为22.5%,55.8%,64.7%;对CYP2E1mRNA表达水平的抑制率分别为36.6%,51.7%,61.4%。结论 漏芦浓度依赖性抑制CYP2E1酶活性,在转录水平下调大鼠肝细胞CY2E1的表达。

     

    Abstract: Objective To study effects of rhaponticum uniflorum(L.)DC on the activity and mRNA expression of CYP2E1.Methods Rathepatocytes sandwich cultures were treated rhaponticum uniflorum(L.)DC in different concentrations(15.6,31.2,62.4 mg/ml)for 48 hour.The enzymic activity of CYP2E1 was reflected the activity of liver microsomal aniline hydroxylase(ANH).The mRNA expression level of CYP2E1 was determined by semi-quantitative reverse transcriptase-polymerase chain reaction(RT-PCR).Results After treatment with the medicine,there was concentration-dependent inhibition in both enzymic activity and mRNA level of CY P2E1 compared with the control.The rates of the inhibitory effect to CYP2E1 activity were 22.5%,55.8% and 64.7%respectively,while those of the inhibitory effect to mRNA level were 36.6%,51.7% and 61.4% respectively.Conclusion Rhaponticum uniflorun(L.)DC inhibits the enzymic activity of CYP2E1 and down-regulates CYP2E1 expression at the transcriptive levels.
