Study on C667T gene polymorphism and susceptibility to esophageal cancer in Kazakh

  • 摘要: 目的 探讨代谢叶酸的亚甲基四氢叶酸还原酶(MTHFR)C677T基因多态性、蔬菜水果摄入量与新疆哈萨克族食管癌的交互作用.方法 采用1:2配比的病例对照研究方法,共调查92例食管癌患者和184名正常对照.应用聚合酶链-限制性片段长度多态(PCR-RFLP)方法检测基因型.结果 在正常对照中,MTHFR677CC、CT、TT基因型频率分别为70.7%,24.5%,4.7%,而在食管癌组中分别为52.2%,43.5%,4.3%(χ2=10.492,P=0.005).等位基因T携带者发生食管癌的危险性为C基因型的1.709倍(95%CI=1.115~2.167).多因素分析发现,携带677C/T基因型的个体发生食管癌的危险性为C/C基因型的7.216倍(95%CI=1.552~33.554);偶尔、经常食用蔬菜水果(叶酸摄入量)均为食管癌的保护因素,OR值分别为0.761(95%CI=0.263~2.202),0.245(95%CI=0.070~0.850).交互作用提示,食用蔬菜水果可降低携带677C/T基因型的个体发生食管癌的危险性.结论 基因-环境因素(叶酸摄入)的交互作用分析提示,叶酸缺乏及MTHFRC677T基因多态性是哈萨克族食管癌发生的危险因素.

     

    Abstract: Objective To study the relationship between esophageal cancer(EC)on Xingjian Kazakh and Methyleneteterahydrofolate reductase(M THFR)C677 T gene polymorphisms.Methods A 1:2 matched case-control study was conducted with 92 cases of exophageal cancer and 184 population-based controls.M THFR genotypes were detected by PCR-RFL P.Results The genotype frequencies of EC group was different from that of controls group(P<0.05).The allele T of M THFR had a 1.709-fold95%CI:1.115~21.67increased risk of developing EC compared with those who had allele C.Multiple Cox regression analysis showed that:the individuals with 677CT genotype had a 7.216-fold95%CI:1.552~33.554increased risk of developing EC compared with those who had 677CC genotype.It could reduce incidence of esophageal cancer to eat fruit often.(O R:0.245;95%CI:0.070~0.850.Interaction analysis showed that taking fruit could reduce incidence of esophageal cancer if the carriers of the M THFR677C/T genotypes.Conclusion The polymprphism of M THFR C677 T was associated with developing EC in Xinjiang Kazakh.This finding suggests gene-environment interaction and is role in the susceptibility to EC,either folate deficiency of M THFR polymorphisms with risk of EC.
