乙醇对小鼠胚胎内皮型一氧化氮合酶表达影响

Effect of ethanol on expression of endothelial nitric oxide synthase in embryonic midbrain of mice

  • 摘要: 目的 探讨乙醇对胚胎中枢神经系统内皮型一氧化氮合酶(eNOS)基因和蛋白表达的影响.方法 应用器官发生期小鼠全胚胎培养模型,8.5d小鼠胚胎以1.0,2.0,4.0和8.0mg/ml乙醇体外染毒48h后,对胚胎中枢神经系统发育进行评分,并以半定量RT-PCR和蛋白印迹(westernblot)方法检测胚胎中脑组织内皮型一氧化氮合酶基因和蛋白表达.结果 随乙醇染毒剂量增加,胚胎头长逐渐减小,中枢神经系统的前脑、中脑和后脑发育评分均逐渐降低,畸形主要表现为神经管闭合不全.乙醇抑制胚胎中脑组织内皮型一氧化氮合酶基因和蛋白表达.结论 乙醇对中枢神经系统的致畸作用可能与诱导胚胎组织内皮型一氧化氮合酶过量表达、造成组织NO含量过多,进而对发育中的胚胎组织造成氧化损伤有关.

     

    Abstract: Objective To invest igate the effect of ethanol on the expression of enothelial nitric oxide synthase(eNOS)in the central nerv ous system(CNS)of embryonic mice.Methods 1.0,2.0,4.0 and 8.0mg/ml ethanol were administrated respectively in vitro to mouse embryos from 8.5d to 10.5d.The development of embryonic CNS was evaluated according to standard scoring system.The mRNA and protein expressions of eNO S were evaluated by RT-PCR and western blot separately.Results The development of embr yonic CN S and the mR NA and protein expressions of eNOS were suppressed by ethanol exposure.Conclusion Suppression of eNOS might play a role in the terato genic action of ethanol in CN S.
