Development of ELISA-kit of quantitative analysis for fumonisin B1

  • 摘要: 目的 研制快速检测粮食中伏马菌素B1(FB1)的试剂盒.方法 在制备抗FB1单克隆抗体基础上,利用间接竞争酶联免疫吸附方法研制FB1快速检测试剂盒.结果 该试剂盒最低检出浓度为5ng/ml,校正曲线线性范围50~500ng/ml,线性方程Y=-0.582X+1.793(r=0.99,P<0.05).与脱氧雪腐镰刀菌烯醇、玉米赤霉烯酮、T-2毒素均无交叉反应.回收率在71.89%~112.95%之间,平均回收率为100.23%.试剂盒常温下可保存225d,实验室内的变异系数和实验室间变异系数均<20%.结论 该试剂盒快速、简单、灵敏,可满足实际工作需要.

     

    Abstract: Objective To develop a rapid,sensitive,quantitative EL ISA-kit for detection of fumonisin B1 in cereals.Methods It was determined by indirect competitive inhibition enzyme-linked immunosor bant assay on the base of monoclonal antibodies against fumonisin B1.Results The limited concentration of detection was 5 ng/ml,linear range was 50~500 ng/ml,the linear equation w as Y=-01 582x+11 793,(r=0.99,P<0.05).There was no cross reaction with deoxynivalenol(DON),zearalenone(ZEN)and T-2toxin.The recovery rate of spiked maize was among 71.89% ~112.95%,and mean recovery rate was 100.23%.The kit can be stored at normal temperature in 225 days,and coefficient of variant within -laboratory and between-laboratory was less than 20%.Conclusion The EL ISA-kit was simple,fast,sensitive and can meet the demand of practical work.
