微波对大鼠大脑皮层及海马突触囊泡蛋白影响

Influence of microwave radiation on synaptic associated proteins in cerebral cortex and hippocampus in rats

  • 摘要: 目的 探讨微波辐射对大鼠大脑皮层和海马突触囊泡相关蛋白如突触素I(synapsinI),囊泡相关膜蛋白-2(vesicle-associated membrane protein-2,VAMP-2)和突触融合蛋白(syntaxin)以及突触囊泡蛋白(synaptophysin)表达的影响。方法 采用30mW/cm2微波辐射Wistar雄性大鼠,于辐射后6h,1,3和7d取材,通过免疫蛋白印迹检测大脑皮层和海马突触体突触素I,VAMP-2,突触融合蛋白和突触囊泡蛋白表达的改变;采用免疫共沉淀检测VAMP-2和突触融合蛋白相互作用的改变。结果 30mW/cm2微波辐射后皮层突触素I于辐射后3d表达减少(P<0.01);海马突触素I表达1d增加,3d减少,7d又增加的波动(P<0.01)。皮层和海马突触囊泡蛋白于辐射后7d表达增加(P<0.01),VAMP-2和突触融合蛋白于辐射后7d内表达均降低。VAMP-2和突触融合蛋白相互作用在皮层于辐射后3d减弱,而海马于辐射后7d内明显减弱(P<0.01)。结论 微波辐射可引起大脑皮层和海马突触囊泡蛋白表达异常,进而影响突触传递功能。

     

    Abstract: Objective To investigate the effect of microwave radiation on expressions of synaptic associated proteins: synapsin I,VAMP-2,syntaxin and synaptophysin.Methods 25 Wistar rats were exposed to microwave which average power density was 30 mW/cm2.Western blot was used to study the change of the expressions of synapsin I,VAMP-2,syntaxin and synaptophysin of synaptosome in cerebral cortex and hippocampus,and immune co-precipitation was used to study the interaction of VAMP-2 and syntaxin.Results It resulted in the decrease of synapsin I at after 3-day radiation in cerebral cortex (P<0.01),but increase at 1 d,decrease at 3 d and increase at 7 d in hippocampus(P<0.01);the increase of synaptophysin at 1~7 d and decrease of VAMP-2 and syntaxin at 6 h~7 d after radiation in cerebral cortex and hippocampus(P<0.01); the decrease of the interaction of VAMP-2 and syntaxin at 3 d in cerebral cortex and at 6 h~7 d in hippocampus after exposure(P<0.01).Conclusion Microwave radiation can induce the abnormity of expressions of synaptic associated proteins in cerebral cortex,and hipoccampus then induce the disorder of synaptic transmission.
