细粒棘球蚴内蒙株疫苗侯选基因克隆及分析

Cloning and sequence analyse of potential vaccine gene of echinococcus granlosus strain in inner Mongolia

  • 摘要: 目的 为确定细粒棘球蚴脂肪酸结合蛋白(FABP)基因的特性并为研究其免疫功能奠定基础。方法 根据已报道的细粒棘球蚴脂肪酸结合蛋白的基因序列(AY024340和AF321119)设计引物,进行FABP基因扩增。结果 成功克隆到细粒棘球蚴内蒙株FABP基因,扩增片段为482bp;序列比对结果显示,细粒棘球蚴内蒙株FABP基因与国内株FABP基因(GenBank登录号为AY024340)序列完全一致,同源性为100%;与国外株FABP基因(GenBank登录号为AF32I119)的同源性为99%。结论 细粒棘球蚴脂肪酸结合蛋白有潜在的抗原表位位点,为研究该蛋白的免疫功能及构建FABP核酸疫苗奠定了基础。

     

    Abstract: Objective To further study the character of echinococcus granulosus fatty acid binding protein(FABP)gene and provide the basis for analysis of its immunological functioning.Methods The specific primers were designed according to published nucleotide sequence of echinococcs granulosus FABP gene in the genbank database(AY024340 and AF321119).The fragment of FABP gene was amplified by PCR.Results The FABP-NM gene was successfully cloned and the full length of the cloned gene was 482bp.The FABP-NM gene showed 100% homology with the Xinjiang strain in China (AY024340)and 99% homology with the foreign strain(AF321119).Conclusion Several possible antigen epitopes located in the FABP,which prov ided the basis for further studying the immunolo gical function of FABP and construct FABP nucleic acid vaccine.
