接振家兔血管内皮细胞bcl-2、bax表达及意义

Expression of bax and bcl-2 in arterial endothelial cells of segmental vibration rabbits and its significance

  • 摘要: 目的 探讨局部接振家兔血管内皮细胞bcl-2、bax基因表达变化及其意义。方法 家兔随机分为低剂量接振组接振强度ahw(4)3.03m/s2、高剂量接振组接振强度ahw(4)12.30m/s2和对照组。2个接振组接振时间为45d,接振后沿后肢股血管分支取相同部位肌肉作病理切片,免疫组化染色,于光镜下观察所取组织的病理变化及凋亡相关基因bcl-2、bax的表达情况。结果 接振组血管内皮细胞破坏性变化,中膜平滑肌增生,管腔内微血栓形成;接振组血管内皮凋亡抑制基因bcl-2表达高于对照组,bax基因表达较对照组明显升高且差异有统计学意义(P<0.01),bcl-2/bax比值较对照组显著降低(P<0.05)。结论 局部振动可导致家兔血管内皮细胞凋亡,2者的变化可能与手臂振动病的发病有一定关系。

     

    Abstract: Objective To study the bcl-2 and bax expression in rabbit exposured to local vibration and its significance.Methods 18 rabbits were divided into three groups atrandom.Two groups were tested by different vibration power and one was normal control group.SABC immunohistochemistry was used to detect the ex pression of bax and bcl-2.Results Light microscopy showed irregular proliferation in the tunica intima and tunica media,especially in the former;spoted smooth muscle cells,proliferation of febrous connective tissue,and degeneration of partial tissues and infiltration of inflammatory cells were observed in the vibration group.Bax expression was significantly higher than that in the normal controls(P<0.05,P<0.01),bcl-2/bax ratio of the cells in the vibration group was significantly lower than that in the controls.Conclusion Segmental vibration may cause the endothelial cell apoptosis;and there may be some relationship between the mechanism of hand arm vibration disease and endothelial cell apoptosis.
