叶酸对高同型半胱氨酸小鼠心脏TGFβ1表达影响

Effect of folic acid on expression of TGF β1 in myocardial of HCY mice

  • 摘要: 目的 研究叶酸(Folic Acid,FA)对高同型半胱氨酸(HCY)小鼠心脏转化生长因子-β1(TGFβ1)表达的影响,探讨心血管疾病防治措施。方法 昆明种小鼠24只,雄性,随机分为3组:正常对照组(NS组,腹腔注射生理盐水0.1ml),高同型半胱氨酸组(HCY组)与叶酸处理组(FA组)腹腔注射HCY16μmol(kg.d)。腹腔注射2h后,NS组和HCY组经口灌胃给生理盐水0.1ml,FA组给FA1.0μmol/(kg·d)连续5d。末次给药后4h,分离脏器。测定小鼠体重、心脏重,常规石蜡切片,免疫组织化学法测定心肌TGFβ1,HPIA-2000高清晰度彩色图文分析系统对心肌细胞面积进行图像分析。结果 与HCY组比较,叶酸组的心脏增重及心/体比值及心肌细胞TGFβ1表达差异有统计学意义(P<0.01和P<0.05),可显著性抑制HCY所引起的心肌细胞面积扩大及乳酸脱氢酶活力过渡增强(P<0.01)。结论 叶酸作为一种细胞的保护因子,可能通过调节TGFβ1的表达而降低高HCY所致的心肌组织损伤,预防心脏疾病发生。

     

    Abstract: Objective To study the effect of folic acid(FA)on the expression of transforming growth factor β1 (TGFβ1) in myocardial cells of Homocysteine(HCY)mice,and to found out the measure to control cardiovascular disease.Methods Twenty four male Kunming mice were randomly divided into three groups:control group(0.1 ml Saline injected in abdominal cavity),HCY group and FA group(HCY 16μmol/kg.d injected in abdominal cavity);Intragastric adrninistration after 2h of abdominal cavity injection:0.1 ml saline in NS group and HCY group,folic acid 1.0μmol/kg·d in FA group.This was done for continous of 5 days.4 h After the last administration,the weight of body and heart were measured,routine paraffin section,the TGFβ1 of cardiac muscle cell were measured with immunohistochemical method.The area of cardiac muscle cell were measured by HPIA-2000 image computer analyzer.Results The weight of heart and heart/body and the expression of TGFβ1 in myocardial cells were significantly decreased(P<0.01,and P<0.05)in FA group than in HCY mice;The augmentation of myocardial cell area and lactic acid dehydrogerlase(LAD)in HCY mice were significantly decreased by folic acid (P<0.01).Conclusion As one of protective agent of the myocardial cell,the folic acid may decrease myocardial cells dam2 age in HC Y mice through adjusting the expression of T GFβ1,and to preventheart disease.
