糖尿病大鼠补锌后新基因片段表达与鉴定

Expression and identification of novel genes differentially expressed in diabetic rats supplemented with zinc

  • 摘要: 目的对糖尿病大鼠补锌后出现的新基因片段进行组织表达与鉴定。方法对前期研究中分离到的补锌糖尿病大鼠与糖尿病对照组比较有异差的基因片段进行克隆、测序、同源性分析后,对发现的6条新基因(Zn-2、Zn-6、Zn-10、Zn-14、Zn-19、Zn-20)进行RT-PCR鉴定,以排除假阳性并观察其在各组大鼠肝脏中的表达水平。结果20条cDNA片段经克隆、测序、同源性分析,发现了6个基因片段(Zn-2、Zn-6、Zn-10、Zn-14、Zn-19、Zn-20),在Gen Bank中未找到高同源性的序列,为新的表达序列标签(EST)。其中4个基因片段在糖尿病对照组、糖尿病补锌组肝脏中的表达量均低于正常对照组,糖尿病补锌组的表达量高于糖尿病对照组(P<0.05)。4个新基因片段被Gen Bank收录,接收号分别为AY952968,AY952970,DQ351838,DQ351839。结论分离并克隆了4条与补锌有关的糖尿病大鼠差异显示的新基因片段,在GenBank上成功登录。

     

    Abstract: ObjectiveTo express and identify novel gene fragments differentially expressed from diabetic rats supplemented with zinc.MethodscDNA fragments from former research project were cloned,sequenced and BLASTn analysed.The RT-PCRof the six novel genes were made to observe the expression changes in liver of various groups.ResultsZn-2, Zn-6,Zn-10,Zn-14,Zn-19,and Zn-20 cDNA were shown to be the novel gene fragments for no matched gene with them in genbank.The expression level of 4 of them,in both DM group and DM+Zn group were obviously lower than those of NC group.The expression level of DM+Zn group was higher than those of DM group(P<0.05).They were registered to genbank with the numbers of AY 952968,AY952970,DQ 351838,DQ351839,respectively.ConclusionThe four novel genes differentially expressed in diabetic rats supplemented with zinc were cloned,and were successfully submitted to genbank.
