五氯酚钠致突变性研究

  • 摘要: 目的研究五氯酚钠的致突变性,为五氯酚钠的毒性机制研究提供依据。方法应用Ames试验、小鼠淋巴瘤细胞试验和体外染色体畸变试验研究五氯酚钠的致突变性。结果五氯酚钠对鼠伤寒沙门菌TA97、TA98、TA100、TA1024株菌株无论直接作用或代谢活化后作用,均未呈现致突变性;随染毒浓度增大,L5178Y细胞tk位点的突变频率有升高的趋势,其中150μg/ml剂量组的突变率与对照组比较,差异有统计学意义(P<0.05);无论是否加入代谢活化剂,受试物均引起染色体畸变率的明显增加。结论五氯酚钠能诱发中国仓鼠卵巢(CHO)细胞染色体畸变,是非基因致突变物或弱基因致突变物。

     

    Abstract: ObjectiveTo investigate the mutagenesis of sodium pentachlotophenol in vitro.MethodsThe mutagenesis of sodium pentachlor ophenol was detected by using Ames test,Mouse Lymphoma Cell Assay(MLA)and chromosome aberration test at different doses.ResultsSodium pentachlorophenol did not induce the reverse mutagenesis of Salmonellaty phimurium TA97、TA98、TA100 and TA102 strain directly or after addition of S9;High dosage sodium pentachlorophenol could incr eased the mutation frequency of TK gene significantly compared with control group.Sodium pentachloro phenol can induce a significantly increased chromosome aberration rate both with and without adding S9 mix.ConclusionSodium pentachlo rophenol can induce chromosome aberration of CHO cell,and is a non-genetic or weak-genetic mutagen.
