植酸对IkB-α表达及胃癌细胞增殖抑制作用

Effect of phytic acid on expressions of gene IkB-α and growth of gastric carcinoma cells

  • 摘要: 目的 研究植酸对抑制性卡巴蛋白(IkB-α)表达影响与抑制胃癌细胞增殖作用机制.方法 采用四甲基偶氮噻唑蓝(MTT)法观察植酸对人胃癌(SGC)7901细胞增殖的抑制作用;倒置显微镜下观察植酸对细胞基本生长状态及形态的影响;单细胞凝胶电泳实验观察植酸对SGC-7901细胞的凋亡诱导作用;蛋白印迹法(Western blot)检测凋亡相关基因IkB-α及可能相关的细胞核因子NF-kBP65蛋白表达.结果 MTT实验结果显示,植酸对SGC-7901细胞具有生长抑制作用,并呈现剂量-时间效应关系;不同浓度的植酸对人SGC-7901 3 d时抑制率分别为26.35%,53.20%,69.29%,86.65%和93.61%.倒置显微镜下的形态学观察表明,加药组细胞的生长状态不佳,植酸作用组细胞出现典型凋亡的彗星脱尾,且存在剂量效应关系,不同浓度的植酸对人SGC-7901 DNA损伤率分别为5.9%,11.0%,34.6%和63.1%.植酸能够上调IkB-α蛋白的表达,植酸处理组细胞中IkB-α蛋白比对照组表达升高,且均呈现剂量-反应关系;同时,植酸能够下调NF-kBP65的蛋白表达.结论 植酸对胃癌SGC-7901细胞具有抑制增殖作用,IkB-α参与了植酸对人胃癌SGC-7901细胞的凋亡诱导作用机制.

     

    Abstract: Objective To explore the effect of phytic acid on apoptosis of gastric carcinoma cells and mechanisms of proliferatory inhibition.Methods The inhibiting action of phytic acid on SGC-7901 cells were examined by MTT assay.The effect of IP6 on mor phology alterations of SG C-7901 cells by reverse discrepancy microscope observation and the effect on apoptosis of SGC-7901 cells was examined by Comet Assay,and the expressions of IkB-and NF-kBP65 protein were detected by Western blot method.Results From MTT assay,phytic acid was observed with the inhibition on the growth of gastric cells in dose and time dependent manners,the rate of cell growth suppression was 26.35%,53.20%,69.29%,86.65% and 93.61% at different concentrations,respectively.Cell growth of dosage group was inhibited.Rate of DNA tail of experimental groups was significantly higher than that of control group,and the rate of DNA damage was 5.9%,11.0%,34.6% and 63.1%.The expressions of IkB-α protein in phyt ic acid group were higher compared with control group,and increased in a dose-dependent manner.Expr essions of NF-kBP65 protein in phytic acid group was lower compared with control group,and reduced in a dose-dependent manner.Conclusion Phytic acid can inhibitit the proliferation of gastric carcinoma SGC-7901 cells which may be correlated to apoptosis associated gene IkB-α.
