大豆苷元对大鼠骨形态学及生物力学影响

Effects of daidzein on histomorphometry and bone biomechanics in rats

  • 摘要: 目的 探讨不同剂量大豆苷元对去卵巢大鼠骨组织形态学和骨生物力学的影响.方法 72只3月龄雌性SD大鼠随机分为6组,每组12只,分别为假手术组(Sham组)和5个卵巢切除术组.术后1周开始灌胃大豆苷元,连续灌胃6个月后处死大鼠,对各组大鼠股骨几何参数进行分析,并进行骨组织形态学观察和骨生物力学的测定.结果 去卵巢组大鼠骨小梁面积、骨小梁厚度、单位面积内的骨小梁数目均呈降低趋势,分别下降25.8%,8.09%,12.4%;骨生物力学性能下降,最大载荷(Fmax)和弹性载荷显著降低(P<0.05).一定剂量大豆苷元>50m/(kg·bw)能对上述指标有所改善,差异有统计学意义(P<0.05).结论 一定剂量大豆苷元>50mg/(kg·bw)对去卵巢骨质疏松大鼠的骨组织形态学和生物力学有改善作用,使其骨量增加,骨结构改善.

     

    Abstract: Objective To investig ate the effects of different dosag es of daidzein on histomorphometr y and bone biome chanics in ovariectomized rats.Methods 72 female SDrats of 3 months were divided into six groups by body weight r andomly.One group was sham group,and the o ther rats in 5 groups were operated to cut out their ov ar ies.Dr enched daidzein was given for 6 months and the r ats were killed.To measure geometr ic parameters,histomorphometry and bone biomechanics of the left femur in differ ent groups of rats were observed.Results The area percent of trabecular,the thickness and the number of tr abecular in OVX group were all reduced with percentage of 25.8%,8.09%,12.4%,respectively.The bone biology mechanics performance dropped,the maximum load and flexibility load dropped obviously.Some dosages of daidzein>50mg/(kg·bw)could antagonize these changes(P<0.05).Conclusion Some dosages of daidzein>50 mg/(kg·bw) can improve the histomor phometry and biomechanics of bone of gelding osteoporosis female rats.
