硫辛酸对氢醌致V79细胞线粒体膜电位影响

Efects of(R)-a-lipoic acid on mitochondrial membrane potential changes in V79 cells caused by hydroquinone

  • 摘要: 目的 利用荧光探针JC-1观察氢醌对V79细胞线粒体膜电位的影响及硫辛酸的保护性作用.方法 在体外培养的V79细胞中分别加入一定浓度的硫辛酸和氢醌,培养24 h,采用倒置荧光显微镜和流式细胞术检测细胞线粒体膜电位水平.结果 正常V79细胞线粒体膜电位为1.12:而50,100μmol/L氢醌可导致V79细胞线粒体膜电位下降,分别为0.22和0.03;50μmol/L的硫辛酸单独作用时不改变线粒体膜电位,但能明显拮抗50,100μmol/L氢醌引起的线粒体膜电位下降,分别为0.79和0.63.结论 一定浓度硫辛酸可有效抑制50,100μmol/L氢醌引起的V79细胞线粒体膜电位的下降,对V79细胞的线粒体膜电位具有保护作用.

     

    Abstract: Objective To explore the changes of mitochondrial transmembrane potential(m)in V79 cells caused by hydroquinone with fluorescent JC-1 and the protection of lipo ic acid.Methods V79 cells were cultured in vitro,then treated with lipo ic acid and hydroquinone into different groups for twenty-four hours.The levels of mitochondrial transmembrane potential were analysed by flowcytometry.Results Normal V79 cells had a high m(1.12),and JC-1 agg regates were for med in the inner membr ane of mitochondria and emitted orange-red fluorescence.50mol/L and 100 mol/L hydroquinone caused the decrease of m,and JC-1 aggr egates were disso ciated to monomer which emitted green fluorenscence(0.22 and 0.03).Treatment with lipoic acid alone didn't change them,but it could effectively protect V79 cells from hydroquinone tox icity(0.79 and 0.63).Conclusion The results showthat lipoic acid can inhibit decrease of mitochondr ial transmembrane potential of V79 cells damaged by hydroquinone.
