茶多酚对内皮细胞粘附分子表达影响

Effect of tea polyphenois on expression of cell adhesion molecules

  • 摘要: 目的 研究茶多酚(Tea Polyphenols,TP)在同型半胱氨酸(Homocysteine,Hcy)诱导人脐静脉内皮细胞(HUVEC)粘附分子表达改变中的作用.方法 将处于对数生长期的HUVEC分为对照组、TP组、Hcy组、TP+Hcy组,采用四甲基偶氮噻唑蓝法(MTT)测定细胞活性,免疫细胞化学方法分析细胞间粘附分子-1(ICAM-1)和E选择蛋白(E-selectin)的表达.结果 MTT结果显示,Hcy可明显抑制HUVEC的生长(36.0%,P<0.05),加入茶多酚后则明显降低Hcy所致的细胞生长抑制(7.9%,P>0.05).免疫细胞化学方法测定结果显示,与对照组比较,Hcy组细胞ICAM-1和E-selectin表达分别升高29.9%和37.7%(P<0.05);茶多酚与Hcy共同培养后,ICAM-1和E-se-lectin表达分别下降了17.9%和24.6%(P<0.05).结论 TP对Hcy诱导的HUVEC粘附分子ICAM-1和E-se-lectin表达改变有一定的逆转作用.

     

    Abstract: Objective To observe the effect of tea polyphenols(TP)on cell adhesion molecules expression of humanum bilicalvein endothelial cells(HU VEC)induced by homocysteine.Methods HU VEC in logar ithm growth per iod were divided into control group and 3 experimental groups,which were TP,Hcy,and Hcy+T P respect ively.The final concentrations of TP and Hcy in medium were 25 mg/L and 5 mmol/L respectively.Control group were treated with equal volume of PBS.After 24 hours administered with TP and/or Hcy,cell viability was determined by methyl thiazolyl tetrazolium(MTT)assay,the expression of inter cellular adhension molecule-I(ICAM-1)and E-selectin were analyzed by immunocy to chemistry.Results MTTresults showed that TP had no effects on HU VEC at 25 mg/L.Hcy inhibited the cell proliferation rate by 36.0%(P<0.05),and T P decr eased the effects of Hcy on HUV EC(7.9%,P<3.05).The results of immunocytoche mistry also showed that the expr ession of ICAM-1 and E-selectin in group of Hcy increased by 29 9% and 37.7%(P<0.05)respectively compared with the control group,while TP could inhibit the effects of Hcy,decreased the expression of ICAM-1 and E-selectin by 17.9% and 24.6%(P<0.05).Conclusion TP could reverse the changes of the expression of ICA M-1 and E-selectin induced by Hcy on HUV EC.
