Expression of mismatch repair gene during different stages of 16HBE cells induced by cadmium chloride

  • 摘要: 目的探讨氯化镉诱发人支气管上皮(16HBE)细胞系恶性转化过程中错配修复基因hMSH2和hMLH1的mRNA和蛋白动态变化情况。方法用反转录一聚合酶链反应(PT-PCR)和免疫组织化学(SP法)检测16HBE细胞、氯化镉恶性转化16HBE细胞系不同阶段(第5,15,35代细胞及成瘤细胞)hMSH2基因和hMLH1基因的mRNA和蛋白的表达情况。结果随着转化代数的增加,hMSH2基因mRNA和蛋白的表达逐渐降低,至第35代细胞后表达明显下降(P<0.01);而hMLHl基因的mRNA和蛋白表达水平在氯化镉恶性转化16HBE细胞过程中无明显变化差异(P>0.05)。结论错配修复基因hMSH2表达水平的下调可能是镉化物分子致癌的重要机制。

     

    Abstract: ObjectiveTo explore the mRNA and protein expression of hMSH2 and hMLH1 gene during different stages of 16HBE cells induced by cadmium chloride(CdCl2).MethodsReverse transcription-polymerase chain reaction(RT-PCR)and immunohistochemcial staining(SP method)were used to measure hMSH2 and hMLHI genes mRNA and protein expression levels in 16HBE cells and their different passage cells treated by cadmium chloride(the 5th,15th,35th passage cells, and tumorigenic cells from nude mice's tumor tissue).ResultsThe bigger the passages times of 16HBE cells treated with cadmium chloride,the lower the expression levels of hMSH2 gene were.The hMSH2 genes mRNA and protein expression levels of the CdCl2 transformed 35LH 16HBE cells and tumorigenic cells of nude mice significantly decreased compared with non-transformed 16HBE cells(P<0.01).There was no significant difference in mRNA and protein expression of hMLH1 gene during different stages of 16HBE cells induced by cadmium chloride(P>0.05).ConclusionThe down-regulated expression levels of hMSH2 gene may be the possible carcinogenic mechanism for cadmium chloride.
