大豆异黄酮对氧化损伤血管内皮细凋亡作用

Inhibitive effects of soybean isoflavone on apoptosis of vascular endothelial cells following oxidative injury in vitro

  • 摘要: 目的探讨大豆异黄酮(SI)对氧化损伤血管内皮细胞凋亡作用。方法体外培养血管内皮细胞,采用氧化低密度脂蛋白(ox-LDL)进行氧化损伤,将细胞分为6组,即空白对照组、氧化损伤对照组(ox-LDL)、氧化损伤加入维生素E对照组(VE+ox-LDL)、氧化损伤加入SI低、中、高剂量组(SI-L+ox-LDL、SI-M+ox-LDL、SI-H+ox-LDL)。将ox-LDL作用于预先加入VE或不同剂量SI孵育24 h的内皮细胞,继续培养24 h,测定细胞四甲基偶氮噻唑蓝(MTT)吸光度(A)值、乳酸脱氢酶(LDH)释放情况、细胞凋亡指数(AI)及形态学观察。结果空白对照组、VE+ox-LDL及(SI-L、M、H)+ox-LDL组细胞MTT(A)明显高于ox-LDL组(P<0.01);空白对照组、VE+ox-LDL及(SI-L、M、H)+ox-LDL组细胞LDH释放百分比及AI均明显低于ox-LDL组(P<0.01)。结论SI对体外氧化损伤血管内皮细胞凋亡具有明显的抑制作用。

     

    Abstract: ObjectiveTo study the effects of soybean isoflavone(SI)on the apoptosis of vascular endothelial cells following oxidative injury in vitro.MethodsThe vascular endothelial cells were cultured in vitro and divided into 6 groups, vascular control,oxidized low density lipoprotein(ox-LDL),VE+ox-LDL,SI-Low(SI-L)ox-LDL,SI-Middle(SI-M)ox-LDL,SI-High(SI-H)ox-LDL grops.The vein endothelial cells were incubated with ox-LDL for 24 hours after incubated with VE on different doses of SI for 24 hours.The levels of MTT(A),LDHreleasing and AI of cells were measured and their morphologic characters were observed.ResultsThe MTT(A)levels of control,VE+ox-LDL and(SI-L,M,H)+ox-LDL groups were significantly higher than those of ox-LDLgroup(P<0.01).The levels of releasing percentage of LDH and apoptosis index in control,VE+ox-LDL and(SI-L,M,H)+ox-LDL groups were significantly lower than those in ox-LDL group(P<0.01).ConclusionSIcan inhibit apoptosis of vascular endothelia cells following oxidative injury in vitro.
