Abstract:
ObjectiveTo observe the expression of cytokine signaling suppressors(SOCSs)mRNA in rats with acute liver failure(ALF).
MethodsThe model of ALF was induced in rats by D-galactosamine/lipopolysaccharide(D-GalN/LPS).Then,the gene expressions of SOCS-1 and SOCS-3 in the liver was determined by RT-PCR,the TNF-α and IL-6 level was estimated by ELISA at 2 h,6 h,12 h,24 h and 48 h after the injection of D-GalN/LPS.
ResultsThe TNF-α and IL-6 level were significantly elevated and reached the peak at 6th hour(
P<0.01).Meanwhile,the gene expressions of SOCS-1 and SOCS-3 were obviously higher than those in the controls and reached the peak at 6th and 12th hour respectively after the injection of D-GalN/L PS(
P<0.01).
ConclusionAcute liver failure can induce the up-regulation of the gene expressions of SOCSs in rats,which closely correlates to the changes of TNF-α and IL-6,indicating that SOCSs may participate in regulating the balance of inflammatory response in acute liver failure.
