间日疟原虫传播阻断疫苗候选抗原Pvs48基因特点

Genetic characteristic analysis of vivax malaria transmission-blocking vaccine candidate antigen Pvs48 in Chinese Yunnan isolates

  • 摘要: 目的 阐明间日疟原虫传播阻断疫苗候选抗原编码基因Pvs48的特点。方法 采集我国疟疾高发混合流行区云南省间日疟患者指尖血样本,制备血样干滤纸片提取疟原虫基因组DNA;聚合酶链反应(PCR)扩增Pvs48;双脱氧链终止法测序,并对测序结果进行序列多态性分析。结果 共成功扩增16例间日疟原虫分离株Pvs48基因。与间日疟原虫标准株Sal-I比较,检测出6处错义突变位点,导致6处氨基酸置换和3种基因型、3种氨基酸型。结论 我国间日疟原虫传播阻断疫苗候选抗原Pvs48编码基因相对保守,不同地理株间差异无统计学意义。有限的基因突变再次提示以Pvs48为基础构建的传播阻断疫苗在我国具有广泛应用的可行性。

     

    Abstract: Objective To investigate the genetic characteristic of vivax malaria transmission-blocking vaccine candidate Pvs48 in Chinese isolates.Methods Blood samples were collected from patients infected with P.vivax from Yunnan epidemic areas.The parasite genome DNA was extracted.The DNA sequences were amplified.PCR products were directly sequenced.Results The 16 sequences of Pvs48 complete gene were obtained.Compared with Pvs48 Sal-I sequence,there were 6 non -synonymous mutation sites,3 kinds of haplotypes and 3 amino acid substitutions.Conclusion The limit genetic mutation of Pvs48 suggests that Pvs48 antigen is conserve.The limit genetic mutation of Pvs48 reinforces that the vivax transmission -blocking vaccine base on Pvs48 would promise a good prospect for a wide range application in China.
