Expression and significance of IL-1α and MMp-2 in herminated lumbar intervertebral disc

  • 摘要: 目的 检测白介素-1α(IL-1α)及基质金属蛋白酶-2(MMP-2)在突出退变腰椎间盘及正常间盘中的表达,分析其在退变腰椎间盘中的作用.方法 收集42例腰间盘突出症手术切除的椎间盘组织,其中突出型22例,游离型20例为实验组;10例正常髓核取自腰外伤骨折患者,经免疫组化染色(SABC法)检测2组IL-1α及MMP-2的表达情况,进行统计学分析.结果 对照组中IL-lα与MMP-2的阳性表达较少或无表达,实验组中IL-1α在在腰椎间盘突出症,突出型阳性率为86.36%,与腰椎间盘突出症游离型阳性率为85%,2者比较差异无统计学意义(P>0.05).而MMP-2在腰椎间盘游离型中阳性率为90%,高于突出型63.63%的表达,差异有统计学意义(P<0.05).结论 IL-1α与MMP2在诱发腰椎间盘退变过程中可能存在一定的关联性.IL-1α及MMP2表达增加可能是导致腰椎间盘退变突出的原因之一.

     

    Abstract: Objective To detect the expression of the IL1 and the M MP2 prote in in lumbar interverbral discand discuss the role of the IL1 and the MMP2 protein in the course of the herniated intervertebral disc.Methods All of samples of herniated lumbardiscs was obtained from 42 adult patients operated.They were divided into two groups;22 cases were protrusion type(group A)and 20 cases were seque stration type(group B).As a control group,10 lumbar discspeci mens were harvested from 8 patients suffering from acute bursting fracture of lumbar of spine Immunohisto chemistry was used to detect expression of the IL1 and the MMP 2 protein.Results The expression level of IL1 and MMP2 was much higher in group A and B than in the control group.The expression of 1L1 showed no significant difference between group A(86.36%)and B(85%),while the expression of MMP 2 was higher in group B(63.63% in group B vs 90% in group A).Conclusion The expression levels of IL1 and MMP2 are higher in degenerative lumbar in tervertebral disc.The up regulation of MMP 2 probably has something to do with the degree of intervertebral disc degeneration,while IL1 seems not related.The increa sing expression of IL 1 and MMP 2 might be one of the reasons the degene ration and prolapse of lumbarintervertebral discs.
