Abstract: Objective To explore the antigenicity of RhDghost cells prepared with using phosphate solution(PBS)at different concentration and the methods to separate anti-Dantibodies from plasm a contatining low titer antibody from the cells.Methods The ORhD positive group red bloodcells(geno type CCD ee)were employed to prep are ghost cells with phosp hate solution at 40,30 and 20 mmol/L and the antigenicity of the cells was examined.The anti-Dantibodies were separated from helthy populationsπs plasm a containing anti-D01814μg/ml through affinity chromatog raphy(AC)with the ghost cells as solid antigen prep a red by PB of 30 mmol/L and the related parameters were examined.Results The ghost cells prepared with pH 4.0,30 mmol/L PB kept complete cell morphological character and effective antigenicity.The anti-D antibodies were separated successfully by A C method from plasma and the recollection rate of anti-D was 84.65%.The content of IgG monomer and dimer together reached 99.3% of total IgG and no IgA,IgM and hemoglobin were found in the preparation.Conclusion The ghost cells prepared with pH 4.0,30 mmol/L PB can be used as solidantigen to separate and puri-fyanti-Dantiboies from plasm a with low titter of anti-D.
