冠心病与热休克蛋白60基因多态性关系

Association of hsp60 gene polymorphisms with coronary heart disease risk

  • 摘要: 目的 研究热休克蛋白60基因单核苷酸多态性rs788016和rs2340690基因型在病例组和对照组中的分布及其与冠心病风险的关系。方法 应用Taqman基因分型方法分别检测674例冠心病病人和774名对照的rs788016和rs2340690基因型分布。结果 rs788016和rs2340690各基因型在病例组和对照组的分布差异无统计学意义。按性别、年龄、吸烟状况进行分层后,在年龄<60岁的男性现在吸烟层中,病例组中Hsp60基因rs788016GG基因型和rs2340690CC+CT基因型的频率低于对照组;rs788016AA+AG相当于GG,OR(95%CI)=1.78(1.04~3.03)为该人群中冠心病发病的危险因素;rs2340690CC+CT相对于TT,OR(95%CI)=0.58(0.62~5.20)。分层分析表明,该基因型和冠心病发病无关。采用Logistic回归模型分析男性吸烟组中rs788016基因型与冠心病的关系,调整其他因素后OR(95%CI)=1.74(1.08~2.82)。结论 在年龄<60岁男性现在吸烟人群中,rs788016AA+AG基因型可以是冠心病发病的危险因素。

     

    Abstract: Objective To investigate the relationship between hsp60 polymorphisms and coronary heart disease (CHD).Methods With Taq Manallelic discrimination method and case-controls tudy,the f requenc ies of rs788016 and rs2340690 in 674 patients with CHD and 744 controls were determined.Results The frequencies of rs788016 and rs2340690 geno types were not different between cases and controls.After adjusting for sex,age and smoking status,the frequencies of rs788016 AA+AG geno type and re2340690 TT geno type in controls were sigificantly higher than that of in patients of male smoker and younger than 60 years.Compared with rs788016 GG geno type,subjects with AA+AG geno type had higher risk of CHD(OR=1.178,95%CI=1.04-3.03).The subjects with rs2340690 CC+CT genotype had similar risk compared with TT geno type(OR=0.58,95%CI=0.62-5.20).The male smoking subjects with rs788016 AA and AG geno type had almost twice of the risk of CHD(OR=1.74,95%CI=1.08-2.82)。Conclusion The genotype of rs788016 may serve as a novel genetic marker for the risk of CHD.
