骨髓间充质干细胞对糖尿病大鼠疗效观察

Therapeutic effect of mesenchymal stem cell on diabetes in rats

  • 摘要: 目的 体外诱导骨髓间充质干细胞(mesenchymal stem cells,MSCs)分化为胰岛样细胞,用于干预治疗糖尿病大鼠。方法 取周龄大鼠骨髓细胞,进行MSC培养纯化扩增,用bFGF、胰岛素等培养体系诱导MSC向胰岛细胞转化;双硫腙染色鉴定胰岛样细胞。使用链脲菌素(Streptozotocin,STZ)制作糖尿病大鼠模型,将诱导分化的胰岛样细胞移植至糖尿病模型大鼠,观察干预治疗效果。结果 体外诱导MSCs由长梭形变成多角形并逐渐聚集成圆形,双硫腙染色后细胞团成亮红色,表明为胰岛样细胞。糖尿病模型大鼠移植诱导分化胰岛样细胞1周后,多饮、多尿症状减轻,10d后,血糖值由(21.352±3.150)mmol/L降低为(14.604±3.236)mmol/L,与对照组(22.256±4.051)mmol/L比较,差异无统计学意义(P>0.05);尿糖值由(18.00±1.00)g/L降低为(8.50±0.25)g/L,与对照组(20.00±0.18)g/L比较,差异有统计学意义(P<0.05)。结论 骨髓间充质干细胞可在体外诱导分化为胰岛样细胞;骨髓间充质干细胞诱导分化的胰岛样细胞可以降低糖尿病大鼠血糖、尿糖值水平,可能具有治疗糖尿病作用。

     

    Abstract: Objective To study the induction of mesenchymal stem cell(MSC)into islet-like cells and its therapeutic effect on diabetes in rats.Methods MSCs were cultured,exp nded in vitro and induced into islet-like cells in high glucose DMEM media with factors like bFGF,EGF.The induced cells were identified by dithizon staining.Differentiated islet-like cells were transplanted into Wistar rats with diabetes mellitus prepared by STZ and the treatment effect was obse rved.Results After the MSCs were induced in vitro,their forms changed from long spindle-shape to polygon or round and attached into islet-like cell.Most of induced cells showed bright red color in dithizon staining.After the differentiated islet-like cells were transplanted into diabetes-model rats for1 week,the syndrome of polydipsia and polyuria was decreased.Ten days later,the blood glucose valuedec reased(14.604±3.326mmol/L)but was not significantly different from that of in control group(22.256±4.051 mmol/L,P>0.05).The urine glucose value decreased(8.50±0.25g/L)and was significantly different from that of control group(20 100±0.18g/L,P<0.05).Conclusion Bone marrow mesenchymal stem cells could beinduced into islet-like cells,and the blood and urine glucose values were decreased after the differentiated cells were transp lanted into the diabetes-model rats.
