巨噬细胞损伤型大鼠矽肺纤维化模型

Silicosis fibrosis model through macrophage injury in rats

  • 摘要: 目的研究巨噬细胞接触SiO2颗粒后,引起巨噬细胞破坏、崩溃的可能性及程度。方法将Wistar大鼠一次性腹腔注入1ml灭菌的SiO2颗粒溶液,第1~3 d分别抽取腹腔液,显微镜下观察巨噬细胞吞噬SiO2颗粒以及巨噬细胞破坏的情况。结果用SiO2颗粒腹腔注入4 h后可以造成巨噬细胞典型的细胞病变、破坏、最后崩溃,并且巨噬细胞的破坏率可达10%以上。结论应用本模型可以研究矽肺纤维化时巨噬细胞在吞噬SiO2颗粒时的病变、信号、信号传导、基因改变、蛋白质变化等。为矽肺致病机制的研究提供了一种简便,客观,容易操作的矽肺模型。

     

    Abstract: ObjectiveTo study the possibiliy and degree of macrophage injury and collapse induced by SiO2.Methods Sterile SiO2 solution(1ml)was injected in to abdominal cavity of Wistar rats one time.The peritoneal fluids of the rats was taken at 1st-3rd day after the exposure to observe the phayocytose and injuries of macrophages.ResultsTypical cytopathic,damage and finally breakdown resulted from the injection of SiO2 particles were observed 4 hours after the injection.The damage rate of macrophage was up to 10%.ConclusionThe animal model of silicos is was established and could be adopted in pathogenic study of silicosis.
