多糖组合优化对小鼠免疫活性影响

Effects of polysaccharide-combinations on immune activity of mice in vivo

  • 摘要: 目的 研究免疫活性最佳的多糖组合和剂量配比,并探讨各多糖间的协同制约关系和综合作用机制.方法 免疫作用靶点不同的4种多糖均设3个剂量,选用L27(313)正交表,形成27种多糖组合,给予小鼠腹腔注射.检测其对免疫器官指数、迟发型变态反应、血清溶血素含量、腹腔巨噬细胞吞噬鸡红细胞活性、脾自然杀伤(NK)细胞活性的影响.结果 免疫活性最佳的多糖组合是地衣多糖8 mg/kg、灵芝多糖16 mg/kg、黄芪多糖8 mg/kg、香菇多糖16 mg/kg;其高、中、低剂量的脾脏指数分别为(1.214±0.153),(1.327±0.168),(1.052±0.074),均明显高于空白对照组的(0.554±0.069)(P<0.05或P<0.01);胸腺指数分别为(0.261±0.065),(0.273±0.049),(0.241±0.053),均明显高于空白对照组(0.140±0.025)(P<0.05或P<0.01).结论 靶点不同的几种多糖以适宜比例组合后,可能以组合方式全面调节免疫的多个作用靶点,产生协同增效作用,从而更好地提高机体免疫.

     

    Abstract: Objective To study the best composition and dose ratio of polysaccha ride-combination in vivo,and to explore the mechanism of its immunoenhancement.Methods This expremental was designed based on the L27(313)o rthogonal.Four polysaccha rides were selected as experimental factors and they were set to three doses from scratch.After polysac charides combination in traperitoneal in jection to the mice,the immune organ index,the spleen nature killer cell activity were studied.Results The best composition of polysaccha ride-combinations on the immune activity was the composition of lichenan(8 mg/kg),ganode ran(16 m g/kg),astraga lan(8 m g/kg)and lentinan(16 mg/kg).In the high,middle,low dosage levels,the index of spleen were 1.214±0.153,1.327±0.168,1.052±0.074,respectively.The index were all higher than the control group(0.554±0.069)significantly(P<0.05 or P<0.01).The index of thymus were 0.261±0.065,0.273±0.049,0.241±0.053,and were all higher than that of the control group(0.140±0.025)significantly(P<0.05 or P<0.01).Conclusion The ploysaccharide-combination with most apropriate doses ratio maybe provide the most effective way for pooducing the strongest immune-enhancing agent.
