甲醛对小鼠源性巨噬细胞DNA损伤作用

Damage effect of formaldehyde on DNA of RAW264.7 cell

  • 摘要: 目的 探讨甲醛对小鼠源性巨噬细胞(RAW264.7细胞)DNA的损伤作用.方法 用0,20,40,80,160,320μmol/L甲醛处理RAW264.7细胞1 h.采用单细胞凝胶电泳技术检测甲醛对RAW264.7细胞DNA的损伤作用.结果 甲醛剂量为20,40,80,160μmol/L时,RAW264.7细胞DNA的迁移率和尾长分别为33.0%,49.7%,82.3%,38.3%和11.2,22.6,30.1,18.9μm,与空白对照组4.3%和9.7μm比较,差异有统计学意义(P<0.05);当甲醛浓度达320μmol/L时,细胞DNA的迁移率和尾长分别为6.3%和9.3μm,与对照组比较,差异无统计学意义(P>0.05).结论 低浓度甲醛可造成RAW264.7细胞DNA的损伤.

     

    Abstract: Objective To explore damage effect of forma ldehyde on DNA of RAW 264.7 cell.Methods Single cell gele lectro phoresis was used for detecting the damagee ffect of forma ldehyde on DNA of RAW 264.7 cel.l The RAW 264.7 cell were exposed to 0,20,40,80,160,and 320 mol/L of formal dehyde for 1hr,respectively.Results It was shown that comet incidence and migration length of DNA of RAW 264.7 cells were increased remarkably in 20,40,80,160 mol/L group.But the cometinc idence and migration length of DNA in 40 mol/L group(49.7%,22.6±18.5 m)were more serious than 160 mol/L group(38.3%.18.9±17.5 m).There was no differrence in comet incidence and migration length between 320 mol/L group and control group.Conclusion The formaldehyde has damage effect on DNA of RAW 264.7 cell.
