水中问号钩端螺旋体基因芯片检测

Detection of Leptospira interrogans in water with DNA chip

  • 摘要: 目的 应用DNA微阵列芯片快速检测水中问号钩端螺旋体。方法 应用基因芯片对不同浓度钩体水、钩体可能生存的不同水体、经水传播的其他病原及疫区可疑水标本进行检测,并以PCR技术为对照,观察基因芯片技术检测钩体的特异性、敏感性及可行性。结果 用芯片和PCR技术对水中不同浓度钩体的检测结果表明,最低检测浓度分别为5和10条钩体/100 mL,芯片检测的敏感性高于常规PCR法;对钩体可能生存的不同生物与理化特性水体(非疫区)及经水传播的其他病原进行检测,结果均为阴性;对疫区20份可疑水标本进行检测,芯片、PCR与培养法检测结果阳性分别为8,5,4份。结论 基因芯片技术可快速、灵敏、特异地检测水中问号钩端螺旋体。

     

    Abstract: Objective To detect Leptospira interrogans in water with the DNA chip.Methods The water with different concentration of Lep to spirainter rogans and other pathogens were detected with DNA chip.The specificity and sensitivity of the DNA chip were observed with compaison of polym erase chain reaction(PCR).Results Positive signals were displayed on the micro chips for water samples with Leptospira interrogans and there was nosignal displayed forwater samples without Leptospira interrogans.The least detection sensitivity of the DNA chip and PCR were 5 and 10 Leptospira interro gans per 100 millmie tres water,respectively.The sensitivity of the DNA chip was higher than that of PCR amplification.The positive numbers of Leptospira in terrogans were 8,5,4 with the DNA chip,PCR and liquid culture,respectively for 20 water samples from epidemic areas.Conclusion Leptospirinterrogans in the water can be detected with the DNA chip more rapidly,specifically and sensitively.
