草苁蓉环烯醚萜苷对肺癌细胞增殖和凋亡影响

Effect of iridoid glucosides from Boschniakia rossica on proliferation and apoptosis of lung carcinoma cell

  • 摘要: 目的探讨草苁蓉环烯醚萜苷(IGBR)对肺癌细胞A549增殖和细胞凋亡的影响。方法采用四甲基偶氮噻唑蓝(MTT)比色法观察IGBR对肺癌A549细胞增殖的影响,采用脱氧核糖核苷酸末端转移酶介导的缺口末端标记法(TUNEL染色法)检测其对A549细胞凋亡的影响,免疫细胞化学法检测凋亡相关蛋白Bax、Bc1-2、P53和Fas蛋白的表达。结果随着IGBR浓度增加和作用时间延长,A549细胞增殖抑制率逐渐升高(P<0.01),作用12,24,48h后,细胞凋亡指数(%)依次为(6.6±0.9),(9.1±1.4),(13.4±1.6);Fas和Bax蛋白表达率分别为(24.5±4.1)%,(11.4±2.7)%;Bc1-2蛋白表达率为(12.5±3.9)%,与对照组(12.7±2.6)%,(28.8±2.0)%,(5.7±3.1)%比较,前2者上升,后者下降,差异均有统计学意义(P<0.01);P53表达率差异无统计学意义(P>>0.01)。结论IGBR可通过诱导肺癌细胞凋亡发挥抗肺癌作用,并具有明显的时间与浓度依赖性。

     

    Abstract: ObjectiveTo study the effect of iridoid glucosides from Boschniakia rossica(IGBR)on proliferation and apoptosis of lung carcinoma cell.MethodsMethyl-thiazolyl-tetrazolium assay was used to examine the proliferation of A549 cell.Terminal deoxynuc leotidyl transferase dUTP nick end labeling method was used for the determination of cell apoptosis and immunohistochemical method was used for the determination of Bax,Bcl-2,P53 and Fas protein expression.ResultsIGBR inhibited the proliferation of lung carcinoma cells in a time-and concentration-dependent manner(P<0.01), and induced apoptosis.Furthermore,IGBR increased the expression of Fas and Bax proteins(P<0.01)and decreased the expression of Bcl-2(P<0.01).While there was no significant change in the levels of P53.ConclusionIGBR could exert an anti-lung cancer effect via induction of cell apoptosis.Its anticancer mechanism needs to be explained.
