三聚氰胺对大鼠肾组织原癌基因表达水平影响

Effect of melamine on mRNA expression of oncogenes in kidney tisssue of rat

  • 摘要: 目的探讨三聚氰胺染毒对大鼠肾组织c-myc,c-fosN-ras基因mRNA表达水平的影响。方法用三聚氰胺对SD大鼠进行亚急性经口染毒,设1个对照组和3个实验组,每天对动物采用经口灌胃染毒1次,染毒时间28d,试验结束时取大鼠肾组织进行荧光定量PCR检测c-myc,c-fosN-ras基因mRNA表达水平。结果雄性大鼠三聚氰胺染毒后c-mycc-fosN-ras mRNA表达水平分别为1.72~3.39,2.34~4.03,2.92~12.08,与对照组比较明显增强(P<0.05,或P<0.01);雌性大鼠三聚氰胺染毒后c-mycc-fosN-ras mRNA表达水平分别为1.56~2.93,2.03~3.52,2.65~10.87,与对照组比较差异有统计学意义(P<0.05,或P<0.01)。结论三聚氰胺亚急性染毒引起大鼠肾组织部分癌基因表达水平升高。

     

    Abstract: ObjectiveTo study mRNA expression of c-myc,c-fos and N-ras oncogenes in kidney tissue of rats administrated with melamine for 28 days.MethodsFive male and 5 female SD rats in every experimental group were administrated with three doses of melamine(low,middle,and high dose)by gavage,respectively,for 28 days.After melamine treatment the animals were sacrificed and the kidneys were taken out for detection of mRNA expression of c-myc,c-fos and N-ras with fluorescent quantitative PCR assay.ResultsMelamine induced a significant increase in mRNA expression of c-myc,cfos and N-ras at low,middle,and high dose.The relative levels of c-myc expression in groups treated with three doses of melamine were 156%-339% higher than that of the control.Meanwhile,c-fos mRNA levels increased by 234%-403% in groups treated with three doses of melamine,and N-ras mRNA levels increased by 265%-1 208% in groups treated with three doses of melamine.ConclusionMelamine could induce a significant increase in mRNA expression of c-myc,c-fos and N-ras in kidney tissue when rats were treated with subacute administration of melamine.The findings indicate that melamine might be a potential carcinogen.
