甲基叔丁基醚对胎鼠脑亚细胞结构及ATP酶影响

Effect of MTBE on ultrastructure and ATPase of brain tissue of fetal mice

  • 摘要: 目的探讨妊娠期接触甲基叔丁基醚(MTBE)对胎鼠脑组织亚细胞结构和磷酸腺苷(ATP)酶活性的影响。方法对40只健康成年昆明种小鼠于整个妊娠期间用MTBE,25,100,400,800mg/(kg·bw)进行灌胃染毒,每天1次,给予食用油作为对照组。测定胎鼠脑组织中ATP酶活性,透射电镜下观察其亚细胞结构。结果孕鼠MT-BE染毒剂量为25,100mg/(kg·bw)时,胎鼠脑组织中各种ATPase活性较对照组均轻微增强(P>>0.05);孕鼠400mg/(kg·bw)染毒时,胎鼠脑组织中Na+,K+-ATPase、Ca2+-ATPase、Mg2+-ATPase活性分别为(5.28±0.24),(5.92±0.50),(2.39±0.21)μmol/(mg·prot),与对照组比较明显降低(P<0.05);透射电镜下可见胎鼠脑组织神经细胞和神经胶质细胞肿胀、变性,胞浆内线粒体和部分细胞膜受损。结论妊娠期母鼠接触MTBE可导致胎鼠脑组织亚细胞结构改变和ATP酶活性降低。

     

    Abstract: ObjectiveTo explore the effect of methyl tertiary-butyl ether(MTBE)on ATPase and ultrastructure of brain tissue of fetal mice during gestation.MethodsForty healthy and pregnant KM mice were distributed in five groups randomly.The MTBE exposure doses were 800,400,100,25 and 0 mg/kg·bw by gavage from the 0 day to the end of gestation,once per day.The levels of ATPase activity were checked and the ultrastructure in the brain tissue of the fetal mice was observed.ResultsCompared with the control,the level of ATPase activity in brain tissue of fetal mice was increased at the exposure doses of 25 and 100 mg/kg·bw(P>0.05).The levels of Na+,K+-ATPase,Ca2+-ATPase,Mg2+-ATPase activity in brain tissue of fetal mice were decreased when the exposure dose was 400 mg/kg·bw(Na+,K+-ATPase=5.28±0.24 μmol pi/mg·prot/h,Ca2+-ATPase=5.92±0.50μmol pi/mg·prot/h,Mg2+-ATPase=2.39±0.21μmol pi/mg·prot/hr, P<0.05).The levels of Na+,K+-ATPase,Ca2+-ATPase dereased significantly when the exposure dose increased to 800 mg/kg·bw(Na+,K+-ATPase=5.01±0.57μmol pi/mg·prot/h,Ca2+-ATPase=5.60±0.37μmol pi/mg·prot/hr, P<0.01).The ultrastructural changes of brain tissue of fetal mice with pregnant mice exposed to MTBE during gestation were edema and degeneration of the nerve cell and neuroglia cell and the damages of mitochondria and cell membrane.ConclusionMTBE could induce subcellular structure changes of brain tissue of fetal mice and decreased the activity of ATPase during gestation.
