Expressions and correlation of MGMT, hMLH1 in esophageal cancer among Kazak patients

  • 摘要: 目的研究2种肿瘤修复基因人O6-甲基鸟嘌呤-DNA甲基转移酶(MGMT)与错配修复基因1(hMLH1)在哈萨克族食管癌患者癌组织及其远癌正常组织中的表达及其与病程特征的关系。方法采用逆转录聚合酶链式反应方法(RT-PCR)检测48例哈萨克族食管癌切除标本中MGMT与Hmlh1 mRNA表达水平,并分析两者与临床病理特征的关系。结果 48例食管癌癌组织中,MGMT mRNA表达阳性42例,阳性率为87.5%;正常组织者中34例,阳性率为70.8%,差异有统计学意义(t=3.032,P<0.05);hMLH1 mRNA表达阳性38例,阳性率为79.2%,正常组织中23例,阳性率为47.9%,差异有统计学意义(t=4.156,P<0.05);MGMT表达与浸润深度、淋巴结转移、肿瘤分期等关系有统计学意义(P<0.05);hMLH1表达与浸润深度、淋巴结转移、分化程度及肿瘤分期等关系有统计学意义(P<0.05);哈萨克族食管癌组织中MGMT与hMLH1表达呈正相关(k=0.560,P<0.05)。结论 MGMT与hM-LH1在哈萨克族食管癌的发生、进展、转移等过程中起到一定作用。

     

    Abstract: ObjectiveTo study expressions and correlation of MGMT and hMLH1 in esophageal cancer tissue and normal tissue among Kazak and to investigate the relationship between their expressions and clinical pathological feature.MethodsWe investigated the expression of the DNA repair enzyme:O6-methylguanine DNA methyltransferase(MGMT), and the mismatch repair(MMR)enzyme hMLH1 in 48 esophageal cancer specimens of Kazak with RT-PCR.The relationship between clinical pathologic feature and the expression of MGMT and hMLH1 mRNA level were studied.ResultsThe positive rates of MGMT and Hmlh1 mRNA level in cancer tissue were higher than those of normal tissues(79.2% vs 47.9%,P<0.05),respectively.And there were significant differences in the expressions of MGMT and hMLH1 protein between different clinical pathological factors such as histologic classification,infiltrating depth,lymph node metastasis,and TNM stages(P<0.05).Kappa analysis indicated that the expressions of MGMT and hMLH1 in the Kazak's esophageal cancer were positively related each other(κ=0.560,P<0.05).ConclusionThe expressions of MGMT and hMLH1 may be related to the carcinogenesis and progression of esophageal cancer among Kazak.
