铅对大鼠星型胶质细胞c-Fos和c-Jun蛋白表达影响

Effect of lead on expressions of c-Fos and c-Jun in rat primarily cultured astrocyte cells

  • 摘要: 目的探讨不同时间点低浓度铅暴露对体外原代培养的星型胶质细胞中即早基因c-Fos和c-Jun蛋白表达的影响。方法出生4~8 d Wistar大鼠脑星型胶质细胞原代培养,1.0μmol/L醋酸铅暴露不同时间后分别收集样品,应用蛋白免疫印迹法测定样品c-Fos和c-Jun蛋白表达的变化。结果 1.0μmol/L醋酸铅处理后,星型胶质细胞中c-Fos和c-Jun蛋白表达均明显降低,与对照组比较,染铅15 min时蛋白表达降低有统计学意义(P<0.05),分别比对照降低33.4%和17.3%;至染铅180 min时c-Fos和c-Jun蛋白表达降至最低,均低于对照的1/2(P<0.01)。结论低浓度染铅可使原代培养的星型胶质细胞中c-Fos和c-Jun蛋白表达持续降低,这可能与铅致大鼠学习记忆功能损害有关。

     

    Abstract: ObjectiveTo observe the effect of low-level of lead on the expression of c-Fos and c-Jun in rat primarily cultured astrocyte cells.MethodsThe primary neural glia cells of Wistar rats of 1-4 days old were cultured.Samples of the cell culture were collected at different time after 1.0μmol/L lead acetate exposure and the contents of c-Fos and c-Jun were assayed with Western blot method.ResultsThe exposure of 1.0μmol/L lead acetate could decrease the expressions of c-Fos and c-Jun remarkably compared with those of the control.There were 33.4% and 17.3% of decrease in c-Fos and c-Jun expression 15 minutes after the exposure compared to those of the control,respectively(P<0.05).The expressions of c-Fos and c-Jun decreased to the lowest 180 minutes after the exposure and were only 1/2 of the control(P<0.01).ConclusionLow-level lead exposure could result in the persistent decrease of the expressions of c-Fos and c-Jun within several hours and the decreases may relate to the damage of learning and memory capacity in rat with lead exposure.
