复方六月青对肝纤维化大鼠血清学指标影响

Effect of compound Liuyueqing on serological indexes in hepatic fibrosis rats

  • 摘要: 目的探讨复方六月青(CLYQ)对肝纤维化大鼠血清学指标影响。方法采用四氯化碳(CCl4)诱导大鼠肝纤维化模型,酶联免疫吸附实验(ELISA)法检测CLYQ对肝纤维化大鼠血清学指标超氧化物歧化酶(SOD)、丙二醛(MDA)、羟脯氨酸(Hyp)、透明质酸(HA)和金属蛋白酶组织抑制因子-1(TIMP-1)的变化。结果与模型对照组比较,高剂量CLYQ组大鼠血清SOD活力(142.59±9.80)U/mL明显升高(P<0.01);高剂量CLYQ组大鼠血清MDA、Hyp、HA和TIMP-1含量分别为(2.28±0.43)nmol/mL、(6.53±0.77)μg/mL、(111.16±23.89)ng/mL和(2180.69±278.02)pg/mL,明显低于对照组(P<0.01);并呈现剂量效应关系。结论 CLYQ通过抑制脂质过氧化和降低胶原合成,发挥抗肝纤维化作用。

     

    Abstract: ObjectiveTo study the effect of compound Liuyueqing(CLYQ)on serological indexes in hepatic fibrosis rats.MethodsHepatic fibrosis model of Sprague-Dawley rats were induced by CCl4.The serum superoxide dismutase (SOD),malondialdehyde(MDA),hydroxyproline(Hyp),hyaluronic acid(HA),and tissue inhibitor of metalloproteinase-1 (TIMP-1)were detected with enzyme linked immunoabsorbent assay(ELISA).ResultsCompared with the model control group,the activity of SOD in the serum was increased significantly in all CLYQ groups(P<0.01)and the serological SOD activity was 142.59±9.80 U/mL in the high dose CLYQ group.The contents of MDA,Hyp,HA and TIMP-1 in the serum decreased significantly in all CLYQ groups(P<0.01 or P<0.05).The serological MDA,Hyp,HA and TIMP-1 contents was 2.28±0.43nmol/mL,6.53±0.77μg/mL,111.16±23.89ng/mL,and 2 180.69±278.02 pg/mL in the high dose CLYQ group,respectively,with dose-effect relationship.ConclusionCLYQ can suppress lipid peroxidation and reduce the collagen synthesis.Hence CLYQ has obvious anti-hepatic fibrosis effect.
