家蝇幼虫血淋巴不同Tricine-SDS-PAGE分析方法比较

Determination of hemolymph of Musca domestica( housefly) larvae with Tricine-SDS-PAGE

  • 摘要: 目的用3种不同的三羟甲基甘氨酸-十二烷基硫酸钠-聚丙烯酰胺凝胶电泳(Tricine-SDS-PAGE)方法分析家蝇三龄幼虫血淋巴。方法分别用不含尿素也不含甘油的分离胶组成的普通Tricine-SDS-PAGE方法,分离胶中有尿素组分的三层胶方法,分离胶中有甘油组分的三层胶方法分析家蝇三龄幼虫血淋巴。结果家蝇幼虫血淋巴中含有丰富的小分子多肽,用不含尿素也不含甘油的分离胶组成的普通Tricine-SDS-PAGE方法不能有效分离分子量在<10.700 kDa的小分子蛋白或多肽;分离胶中有尿素组分的三层胶方法虽能清晰分离家蝇幼虫淋巴中<16.949kDa的小分子蛋白或多肽,但对其分子量的测定可能不够准确;而分离胶中有甘油组分的三层胶方法既能很好分离>14.4 kDa的蛋白,也能清晰分离<8.159 kDa的小分子多肽,并能较准确的反映其分子量大小,用于分析家蝇幼虫血淋巴效果较好。结论分析家蝇幼虫血淋巴应采用分离胶中有甘油组分的三层胶的电泳方法。

     

    Abstract: ObjectiveThree different methods of Tricine-sodium dodceyl sulfate polycarylamide gel electrophoresis (Tricine-SDS-PAGE)were used to analyze the hemolymph of Musca domestica(housefly)larvae.MethodsThe hemolymph of Musca domestica(housefly)larvae was analyzed with the separating gel without urea or glycerol in ordinary Tricine-SDS PAGE method,with the urea separating gel method,and the separating gel with glycerol method.ResultsThe results showed that there were abundant low molecular peptides in the hemolymph of housefly larvae.The separating gel without urea or glycerol in ordinary Tricine-SDS PAGE method could not effectively separate the low molecular proteins and peptides with the molecular weight below 10.700 kDa.The result of analysis with the urea separating gel showed that it could effectively separate the hemolymph of housefly larvae,especially the low molecular peptides,but the determination of its molecular weight was not accurate.And the separating gel with glycerol achieved good results not only in separating the low molecular peptides but also in determining the molecular weight.ConclusionThe method of the separating gel with glycerol could achieve satisfactory results in the determination of the hemolymph of housefly larvae.
