黄绿青霉素生物合成及对心肌和血清酶影响

Biosynthesis of CIT and damage of myocardial structure and function induced by CIT in rats

  • 摘要: 目的进行黄绿青霉素(citreoviridin,CIT)生物合成和纯化,并观察其对大鼠心肌组织结构和功能的影响。方法取市售玉米接种黄暗青霉菌孢子后,低温培养3周,霉变米粉经乙醇、苯、正己烷提取后,通过硅胶柱,形成高纯度黄绿青霉素。用CIT15 mg/(kg.d)污染饲料喂养大鼠8周后,取其心肌,进行光学和电子显微镜观察;采用全自动生化分析仪检测血清中谷草转氨酶、谷丙转氨酶、碱性磷酸酶、乳酸脱氢酶、肌酸激酶及其同工酶等,化学发光法检测血清中肌钙蛋白T含量。结果最终合成的黄绿青霉素成分单一,所含杂质较少,并可引发大鼠心肌变性和坏死,病变呈灶状或条索状分布,涉及面较大,有大量炎性细胞浸润,间质水肿,肌浆凝聚,肌原纤维凝集崩解,部分心肌细胞颗粒变或空泡变性;电子显微镜下可见,实验组心肌细胞核固缩呈凋亡状态,核周大量肌丝溶解,肌丝结构明显紊乱,Z线增粗,线粒体数量减少;该毒素还可使大鼠血清中乳酸脱氢酶、谷草转氨酶、肌酸激酶及其同工酶、肌钙蛋白T含量增多,其差异存在统计学意义。结论CIT可使大鼠心肌发生变性和坏死。

     

    Abstract: ObjectiveTo biosynthesize and purify citreoviridin(CIT)and to observe the effects of CIT on the myo-cardial structure and function in rats.MethodsAfterino culation with P.citreon igrum dierckx spores,the commercialminor ground corn were cultivated for 3 weeks.Mouldy corn powder was extracted with ethanol, hot benzene and n-hexane,respectively.The deposit was added to silica gel column with n-haxane-acetone(1:1)as solvent.The yellow fraction was dissolved in methanol to obtain the yellow powder.Rats in treated group were fed with food contaminated with CIT at 15 mg/kg·d for 8 weeks.The hearts of the rats were rapidly removed,and small blocks of myocardium were fixed for light and electron microscopy examination.The content of serumtropon in Tand serum activities of alanine amino transferase,a sparta teamino transfe rase,alkaline phosphatase,lactate dehydrogenase,creatine phosphokinase and its isozyme were detected.ResultsThe final synthesis of CIT obtained a single component with minor impurity.The effects of CIT on the myocardial structure and function in rats were observed.The prominent histopathological changes induced by CIT were manifested as extensive infiltration of lymphocytes and mononuc lear cells,interstitial edema,sarco plasmic condensation,myofibrillar disintegration,and cardiacmyocyte granular degeneration or vacuolation.Electron microscopy confirmed that the pyknotic nucleus presented in apoptosis state;myofilamentlysis and incrassate Z band were observed;mitochondria were reduced in the surrounding of thnucleus.Serumactivities of lactate dehydro genase,aspartateamino transferase,creatine phosphokinase and its isozyme as well as the content of tropon in T were significantly increased compared to those of the control.ConclusionThe synthetic method used in the study is simple,easy,and suitable for laboratory research.CIT could cause degeneration and necrosis in ratmyo-cardium.
