锌对慢性酒精中毒肝损伤再生影响

Effects of zinc supplementation on liver regeneration in mice exposed to long-term ethanol administration

  • 摘要: 目的 探讨锌对慢性酒精中毒肝损伤再生的影响.方法 健康成年雄性C57BL6小鼠40只,随机分为对照组、慢性酒精中毒、酒精中毒+锌及对照+锌4组,持续喂养6个月,形成慢性酒精中毒模型性肝病.测定血清和肝组织中锌和铁含量以及谷丙转氨酶(ALT)活性,免疫组织化学检测增殖细胞核抗原(PCNA)阳性细胞,逆转录-聚合酶链反应(RT-PCR)方法测定肝细胞核4α因子(HNF-4α)mPNA的表达变化.结果 慢性酒精中毒小鼠肝组织锌含量为(6.83±0.82)μmol/L,明显减少,低于对照组的(15.71±1.63)μmol/L(P<0.01);与对照组ALT活性(26.2±1.4)U/L比较,慢性酒精中毒组ALT活性(60.5±16.8)U/L升高(P<0.05);补锌进一步增加了PCNA阳性细胞的数目,HNF-4 mRNA水平因酒精而明显减弱.结论 补充锌能够增强慢性酒精中毒小鼠肝再生,其机制与肝细胞核4α因子(HNF-4α)的增加有关.

     

    Abstract: Objective To examine whether dietary zinc supplemen tation could improve liver regeneration in a mouse model of a lcoholic liver disease.Methods Adult C57BL 6 mice were fed with ethanol-containing liquid diet for 6 months to induce a lcoholic liver disease ascertained by serumalanine tran sfera seactivity and histopathological changes.At the same time, the number of proliferation nuclear antigen(PCNA)-positive cell and the expression of hepa to cyte nuclear factor-4a(HNF-4a) were measured.Results Zinc supplementation to ethano-l exposed mice enhanced liver regeneration by increasing the number of proliferation nuclear antigen(PCNA)-positive cell.Zinc-enhanced liver regeneration was associated with an increase in HNF-4a.Conclusion The studyprovides evidence that zinc supplementation enhances liver regeneration at least in part by HNF-4a through the up-regulation of cell proliferation-related proteins, suggesting that dietary zinc supplementation may have beneficial effects in a lcoholic liver disease.
