Expression of Wnt5a in renal tissue of diabetic rats and its significance

  • 摘要: 目的 探讨Wnt5a基因及蛋白在糖尿病大鼠肾组织中的表达及意义。方法 链脲佐菌素(streptozotocin,STZ)诱导大鼠糖尿病模型,分为2、4、8、12、16、24周组,每一时间组均设正常对照,测定各组血糖、24 h尿蛋白、血肌酐水平。免疫组化方法检测Wnt5a及FN蛋白表达,蛋白免疫印迹(western blotting)检测Wnt5a蛋白表达,逆转录-聚合酶链反应检测Wnt5a及FN基因表达。结果 与正常对照组比较,糖尿病组Wnt5a蛋白表达分别增多了65.5%、72.2%、60%、68.8%、68.8%及63.0%(P<0.01);糖尿病各组Wnt5a和FN mRNA的表达分别增多了50.9%、50%、35.7%、50.9%、55%、54.2%和35.5%、64.6%、34.1%、61.8%、66.7%、64.6%(P<0.01);Wnt5a与FN mRNA表达水平呈正相关(r=0.714,P<0.01)。结论 Wnt5a蛋白和基因在糖尿病大鼠肾组织表达增多,Wnt5a可能参与了糖尿病的发生发展过程。

     

    Abstract: Objective To explore the expression of Wnt5a in renal tissue of diabetic rats.Methods The diabetic rats induced by streptozotoc in(STZ) were randomly divided into 2, 4, 8, 12, 16, and 24 weeks groups and each group was set a case matched control group Blood glucose, 24 hour urine protein, serum creatinine(Scr) of the rats were measured.The prote in and mRNA expression of Wnt5a and fibronectin(FN) in renal cortex were detected with immunoh istochemical staining and reverse tran scriptase-PCR (RT-PCR).Western blotting was employed to detect the expression of Wnt5a protein.Results The Wnt5a prote in could not be detected by immunohisto chemical staining in normal renal tissues.Compared to the control group, the expression of Wnt5a prote in were highly increased by 65.5%, 72.2%, 60%, 68.8%, 68.8%, and 63.0% (P<0.01) by western blotting in diabetic rats.The mRNA of Wnt5a and FN were significantly up regulated by 50.9%, 50.0%, 35.7%, 50.9%, 55.0%, and 54.2% and 35.5%, 64.6%, 34.1%, 61.8%, 66.7%, and 64.6% (P<0.01) in the diabetic rats compared with those of the controls.The expression of Wnt5a mRNA was positively correlated with FN in diabetic rats (r=0.714, P<0.01).Conclusion The findings suggest that Wnt5a gene and prote in expression are up-regulated in the kidney of diabetic rats,which may involve in the pathogenesis of diabetic nephropathy.
