草苁蓉多糖红细胞保护作用

Protective effects of polysaccharides from Boschniakia rossica on erythrocyte

  • 摘要: 目的 研究草苁蓉多糖(BRPS)的红细胞保护作用.方法 常规方法提取BRPS,以硫酸亚铁-水杨酸法测定BRPS对羟基自由基(·OH)的清除能力;以硫代巴比妥酸(TBA)法检测BRPS对红细胞抗脂质过氧化的影响,以分光光度法检测BRPS对红细胞溶血的影响.结果 BRPS具有清除羟基自由基能力,浓度小于0.25 mg/mL时,回归方程为y=170.8x+9.5645,R2=0.993;呈明显浓度依赖性,其对·OH的半数清除浓度为0.24 mg/mL;BRPS明显抑制红细胞脂质过氧化作用和减少红细胞溶血程度,其对·OH诱导的红细胞及纯化红细胞膜体系脂质过氧化作用的半数抑制浓度分别为0.047和0.112 mg/mL,对·OH诱导的红细胞溶血的半数抑制浓度分别为0.068 mg/mL.结论 BRPS具有抗脂质过氧化和红细胞保护作用.

     

    Abstract: Objective To study protective effect of Boschniakia rossica polysaccharides(BRPS) on erythrocyte.Methods BRPS was extracted by routine methods and the ·OH scavenging capacity was examined with FeSO4-salicylic acid method.The effect of BRPS on lipid peroxidation of erythrocytes was estimated by thiobarbituric acid(TBA) method and the effect on hemolysis of erythrocytes was detected by spectrophotometric method.Results BRPS exhibited a·OH scavenging capacity in a concentration-dependent manner with a regeression equation of y=170.8x+9.5645,R2=0.993,and the concentration of inhibitor,which causes 50% inhibition(IC50),of 0.24 mg/mL.BRPS inhibited ·OH-induced lipid peroxidation of erythrocytes and purified erythrocyte membrane with the IC50s of 0.047 and 0.112 mg/mL,respectively,and suppressed the hemolysis of erythrocytes induced by ·OH with the IC50 of 0.068 mg/mL.Conclusion BRPS has anti-lipid peroxidative and protective effects on erythrocyte.
